摘 要:目的探讨侧柏叶提取物对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)增殖、迁移和血管生成的影响,为侧柏叶治疗雄激素性脱发(androgenetic alopecia,AGA)提供新的理论依据。方法取对数生长期的HUVECs细胞株,分别用不同梯度浓度的侧柏叶提取物(30、100、300、1000、3000、9000μg/mL)处理细胞24小时,通过CCK-8实验,检测侧柏叶提取物对HUVECs细胞的安全有效浓度。将细胞分为对照组与侧柏叶组,采用CCK-8方法评估在最佳有效浓度下,侧柏叶提取物处理24、48、72小时后HUVECs细胞的增殖活性。采用细胞划痕实验,观察侧柏叶提取物处理24小时后细胞的迁移能力。采用免疫荧光染色与免疫印迹(Western blot,WB)法检测侧柏叶提取物对HUVECs细胞中血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)与成纤维细胞生长因子2(fibroblast growth factor 2,FGF2)蛋白的表达。结果100μg/mL的侧柏叶提取物对细胞有明显促增殖作用,浓度为300μg/mL时开始出现细胞毒性。与对照组相比,侧柏叶组干预HUVECs细胞24、48、72小时后,HUVECs细胞增殖率均升高(P<0.05),48小时最为显著。与对照组比较,干预24小时后,侧柏叶组中细胞迁移能力升高(P<0.05),且VEGFA与FGF2蛋白的表达水平升高(P<0.05)。结论侧柏叶提取物可以促进HUVECs细胞的增殖、迁移,并可以升高HUVECs细胞中VEGFA和FGF2的表达水平,这可能为侧柏叶在AGA治疗中的分子机制提供了新的理论依据。Objective Exploring the effects of extracts from Platycodon orientalis on the proliferation,migration,and angiogenesis of human umbilical vein endothelial cells(HUVECs),and providing new theoretical basis for the treatment of androgenic alopecia(AGA)with Platycodon orientalis.Methods The HUVECs in the logarithmic growth phase was obtained,and the cells were treated with a range of concentrations of Platycodon orientalis leaves extract(30,100,300,1000,3000,and 9000μg/mL)for 24 hours.The safe and effective concentrations of the Platycodon orientalis leaves extract on HUVECs was determined by CCK-8 assay.The cells were divided into the control group and the Platycodon orientalis leaves group,and the CCK-8 assay was used to evaluate the proliferation activity of HUVECs treated with Platycodon orientalis leaves extract at the optimal effective concentration for 24,48,and 72 hours.A cell scratch assay was employed to observe the migratory ability of cells after treatment with Platycodon orientalis extract for 24 hours.The methods of immunofluorescence staining and Western blot(WB)were used to detect the expression of vascular endothelial growth factor A(VEGFA)and fibroblast growth factor 2(FGF2)proteins in HUVEC cells by the extract of Platycodon orientalis leaves.Results The extract of Platycodon orientalis at a concentration of 100μg/mL was found to promote cell proliferation significantly,while cytotoxicity began to appear at a concentration of 300μg/mL.Compared with the control group,the proliferation rate of HUVEC cells was increased after treatment with Platycodon orientalis leaf extract at 24,48,and 72 hours(P<0.05),with the most significant was increased observed at 48 hours.Compared with the control group,the cell migration ability was enhanced after 24 hours of intervention(P<0.05),and the expression levels of VEGFA and FGF2 proteins in the Platycodon orientalis group were higher than those in the control group(P<0.05).Conclusion The proliferation and migration of HUVEC cells can be promoted by the extract
