基于抗氧化应激探讨侧柏双参方对雄激素性脱发小鼠促生发的效果

摘  要:目的观察侧柏双参方对雄激素性脱发(androgenetic alopecia,AGA)模型小鼠的干预效应,基于氧化应激环节探讨其潜在作用机制。方法60只雄性C57BL/6J小鼠随机分为空白组、模型组、米诺地尔组及侧柏双参低、中、高剂量组。小鼠脱毛后,除空白组外,其余小鼠均外擦睾酮溶液(5 mg/mL)建立AGA小鼠模型,米诺地尔组外抹5%米诺地尔溶液,侧柏双参低、中、高剂量组分别给予的外涂浓度为50、100、200 mg/mL,各组小鼠均每日给药1次,干预16天,分别观察不同时间点小鼠毛发生长速度。通过拍照、皮肤毛发观察仪、皮肤颜色检测、组织苏木素—伊红(hematoxylin-eosin,HE)病理染色观察小鼠毛发再生及毛囊周期转变情况。采用氧化应激试剂盒检测各组小鼠皮肤组织中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)水平。结果干预第9、16天,与模型组相比,米诺地尔组、侧柏双参低、中、高剂量组均能不同程度促进小鼠毛发生长(P<0.05),高剂量效果最为显著;干预第9天,与模型组比较,侧柏双参低剂量组小鼠皮肤组织GSH水平升高(P<0.05),SOD、MDA水平无明显差异,侧柏双参中、高剂量组皮肤组织SOD、GSH水平升高,MDA水平下降(P<0.05);干预第16天,与模型组比较,侧柏双参低剂量组皮肤组织MDA水平下降,GSH水平升高,SOD水平无明显差异,中、高剂量组皮肤组织SOD、GSH水平升高,MDA水平下降(P<0.05)。结论侧柏双参方可以促进AGA模型小鼠毛发再生,其机制可能与抗氧化应激相关。Objective Observing the intervention effects of Cebai Shuangshen Formula on androgenic alopecia(AGA)model mice,and exploring their potential mechanisms of action based on oxidative stress.Methods Sixty C57BL/6J mice were randomly divided into six groups:the blank group,the model group,the minoxidil group,and the three dosage groups of the herbal formula at low,medium,and high doses.After depilation of mice,except the blank group,all the other mice were treated with an external application of testosterone solution(5 mg/mL)to establish an AGA mouse model.The minoxidil group was treated with an external application of 5%minoxidil solution,and the low,medium,and high dose groups were given external application concentrations of 50,100,and 200 mg/mL,respectively.Each group of mice was administered once a day for 16 days of intervention,and the hair growth rate of the mice was observed at different time points.The regeneration of mice hair and the transition of hair follicle cycles were observed through photographic documentation,dermatoscopy,skin color measurement,and histological examination with hematoxylin and eosin(HE)pathological staining.The levels of superoxide dismutase(SOD),malondialdehyde(MDA),and glutathione(GSH)in the skin tissues of mice in each group were detected using an oxidative stress test kit.Results Compared with the model group,on the 9th and 16th day of intervention,the minoxidil group,as well as the low,medium,and high-dose groups of the formula,were able to promote hair growth in mice to varying degrees(P<0.05),with the high-dose group showing the most significant effect;On the 9th day of intervention,compared with the model group,the low-dose group showed an increase in GSH levels in the skin tissue of mice(P<0.05),while there was no significant difference in SOD and MDA levels.The medium and high-dose groups showed an increase in SOD and GSH levels in the skin tissue and a decrease in MDA levels(P<0.05);On the 16th day of intervention,compared with the model group,the low-dose group showed a
