白芍总苷对瘀热互结证发热大鼠炎症因子及血管活性物质的影响

摘  要:目的研究白芍总苷对瘀热互结证发热大鼠的影响及其作用机制。方法将60只雄性SD大鼠适应性喂养后,按体质量随机分为六组,分别为正常组、模型组、阳性药组、白芍总苷低剂量组、白芍总苷中剂量组、白芍总苷高剂量组,每组10只。白芍总苷各剂量组予以不同浓度白芍总苷溶液腹腔注射,给药容积为10 mL/kg;正常组和模型组给予同体积生理盐水腹腔注射,每天1次,连续7天;给药第4天,除正常组外,其余组别大鼠经腹腔注射角叉菜胶50 mg/kg,每天1次,连续3天;同时阳性药组予以阿司匹林灌胃(100 mg/kg),连续3天;末次注射角叉菜胶24小时后,于大鼠背部皮下注射20%干酵母混悬液10 mL/kg,造成瘀热互结证发热大鼠。干酵母混悬液皮下注射6小时后,记录体温,并给予不同药物干预。给药30分钟后再次记录体温,随后将大鼠麻醉,使用数码相机对大鼠的舌面、舌底、爪甲、尾巴等部位进行图像采集,并使用“大鼠体征采集分析系统”对图像进行数据化分析。体征采集结束后,腹主动脉取血和下丘脑组织,采用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)技术检测大鼠血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、干扰素-γ(interferon-γ,IFN-γ)、内皮素-1(endothelin-1,ET-1)、血栓素(thromboxane,TXB2)、6-酮前列腺素F1α(6-oxoprostaglandin F1α,6-keto-pgf1α)含量;采用比色法测定血清一氧化氮(nitric oxide,NO)含量;ELISA技术检测大鼠下丘脑中环磷酸腺苷(cyclic adenosine monophosphate,CAMP)、前列腺素E2(prostaglandin E2,PGE2)含量;采用发色底物法测定大鼠凝血四项。结果与正常组相比,模型组大鼠体温升高,舌面、舌底、爪甲、尾巴红绿蓝(RGB)值与正常组有显著差异(P<0.05)。下丘脑CAMP、PGE2和血清中TNF-α、IL-6、IL-1β、IFN-γ各指标表达均升高(P<0.05);大鼠�Objective To study the effect of Paeonia lactiflora total glucoside on rats with fever with stasis-heat interconjugation syndrome and its mechanism of action.Methods After acclimatisation feeding,60 male SD rats were randomly divided into the normal group,the model group,the positive drug group,the Paeonia lactiflora total glucoside low dose group,the Paeonia lactiflora total glucoside medium dose group,and the Paeonia lactiflora total glucoside high dose group.The total paeoniflorin dose group was given intraperitoneal injections of different concentrations of total paeoniflorin solution,with an administration volume of 10 mL/kg;the normal group and the model group were given intraperitoneal injections of the same volume of physiological saline,both of which were given once a day for 7 consecutive days.On the 4th day of drug administration,except the normal group,the rats in the other groups were injected intraperitoneally with carrageenan gum 50 mg/kg once a day for 3 consecutive days.At the same time the positive drug group was given intraperitoneal injection of Danshen injection at a volume of 10 mL/kg for 3 consecutive days.Twenty-four hours after the last injection of carrageenan gum,20%dry yeast suspension 10 mL/kg was injected subcutaneously into the back of rats,causing stasis-heat interconjugation evidence in febrile rats.Six hours after subcutaneous injection of dry yeast,body temperature was recorded and different drug interventions were given.After 30 minutes of drug administration,the body temperature was recorded again,and then the rats were anaesthetized,and the images of the tongue surface,tongue base,paw nails,tail and other parts of the rats were captured using a digital camera,and the images were analysed using the“Rat Signs Capture and Analysis System”.After the collection of signs,blood was collected from the abdominal aorta and hypothalamus tissues,and enzyme-linked immunosorbent assay(ELISA)was used to measure the levels of TNF-α,IL-6,IL-1β,IFN-γ,ET-1,TXB2 in serum,and 6-keto-PGF1a;
