摘 要:目的 探讨中药毓麟珠对自然衰老大鼠沉默信息调节因子蛋白1(sirtuins 1,SIRT1)/端粒酶逆转录酶(telomerase reverse transcriptase, TERT)的影响并分析其改善卵巢功能的机制。方法 选用24只雌性SD自然衰老大鼠,采用随机数字表法分为衰老给药组和衰老空白组,每组12只;另用6只育龄期雌性SD大鼠为青年对照组。衰老给药组大鼠予毓麟珠水煎液灌胃(10.08 g/kg),衰老空白组予等量生理盐水灌胃,青年对照组正常饲养,连续给药6周后,取大鼠血清和卵巢。苏木精—伊红染色观察卵巢组织形态变化;酶联免疫吸附测定法检测血清抗苗勒管激素水平;化学发光法检测卵巢组织中超氧化物歧化酶、还原型谷胱甘肽(glutathione, GSH)、丙二醛的含量;实时荧光定量PCR法检测卵巢组织中端粒酶活性;免疫组织化学法检测卵巢组织中促卵泡激素受体(follicle-stimulating hormone receptor, FSHR)和雌激素受体(estrogen receptor, ER)表达;蛋白质免疫印迹法检测卵巢组织中SIRT1、TERT蛋白的表达。结果 与衰老空白组相比,衰老给药组卵巢组织中各级卵泡数量增多,形态规则,闭锁卵泡数量减少;血清抗苗勒管激素水平均降低(P<0.05);卵巢组织中GSH、超氧化物歧化酶水平升高(P<0.05),丙二醛水平降低(P<0.05);端粒酶活性升高(P<0.05);促卵泡激素受体、雌激素受体表达明显上调(P<0.05);SIRT1、TERT蛋白的表达均有增加(P<0.05)。结论 毓麟珠可通过影响SIRT1及TERT的蛋白表达,改善自然衰老大鼠的卵巢微环境以及卵巢储备,进而延缓卵巢衰老。Objective To explore the effect of Yulinzhu,a traditional Chinese medicine,on SIRT1/TERT in naturally aging rats and analyze its mechanism for improving ovarian function.Methods Twenty-four female SD naturally aging rats were selected and randomly divided into the aging treatment group and an the aging blank group using a random number table method,with 12 rats in each group.Another six female SD rats of reproductive age were used as the young control group.Rats in the aging treatment group were gavaged with a medium dose of Yulinzhu(10.08 g/kg),while rats in the aging blank group were gavaged with the same volume of normal saline.The young control group was maintained under normal conditions.After 6 weeks of continuous administration,serum and ovaries were collected.Hematoxylin-eosin(HE)staining was used to observe the morphological changes of ovarian tissues.The enzyme-linked immunosorbent assay(ELISA)was used to detect the level of anti-Müllerian hormone(AMH)in serum.The chemiluminescence method was used to detect the contents of superoxide dismutase(SOD),glutathion(GSH),and malondialdehyde(MDA)in ovarian tissues.Real-time fluorescent quantitative PCR was used to detect telomerase activity in ovarian tissues.Immunohistochemistry was used to detect the expressions of follicle-stimulating hormone receptor(FSHR)and estrogen receptor(ER)in ovarian tissues.Western blotting was used to detect the expressions of silent information regulator protein 1(sirtuins1,SIRT1)and telomerase reverse transcriptase(TERT)proteins in ovarian tissues.Results Compared with the aging blank group,the number of follicles at all levels in the ovarian tissue of the aging treatment group were increased,the morphology was regular,and the number of atretic follicles were decreased.The level of AMH in serum was decreased(P<0.05).The levels of GSH and SOD in ovarian tissues were increased(P<0.05),and the level of MDA was decreased(P<0.05).Telomerase activity was increased(P<0.05).The expressions of FSHR and ER were significantly up-regulated(
