摘 要:为了初步研究猕猴桃生长素诱导基因AcIAA9编码蛋白结构及其在猕猴桃生长发育过程中的功能,以‘红阳’猕猴桃为材料,利用PCR技术克隆获得AcIAA9基因全长开放阅读框为1152bp,编码383个氨基酸,疏水系数为-0.539,具有较高的亲水性氨基酸比例,可知AcIAA9蛋白为亲水性蛋白;预测其分子质量为41.04ku,等电点为8.45,不稳定系数为57.31;此外,AcIAA9蛋白具有Aux/IAA家族蛋白典型结构域和保守序列;亚细胞定位预测分析表明,AcIAA9蛋白定位于细胞核内;进化分析表明,猕猴桃AcIAA9基因与茶树CsIAA9基因同源性最高,与葡萄VvIAA9基因位于同一进化分支。该基因启动子区含有多样化的顺时作用元件,除参与调控真核基因转录的TATA框、CAAT框等基本元件外,还具有多种逆境胁迫响应因素,如厌氧胁迫、干旱胁迫等。Real-timeqPCR结果显示,AcIAA9基因在不同组织中存在表达差异,而花中表达量最高。To investigate the protein structure and function of AcIAA9 in the development of kiwifruit.the open reading frame(ORF)of the AcIAA9 gene with a full length of 1152 bp was obtained by PCR from‘Hongyang’kiwifruit.The AcIAA9 gene encoded a total of 383 amino acids.The hydrophobic coefficient was-0.539,with a high proportion of hydrophilic amino acids,indicating that AcIAA9 protein was a hydrophilic protein.The AcIAA9 protein molecular mass was 41.04 ku,the isoelectric point 8.45,and instability coefficient 57.31.The amino acid sequence alignment showed that AcIAA9 possessed a typical structure domain and conservative basic sequence of Aux/IAA gene families.Subcellular localization analysis indicated that AcIAA9 was located in nuclear.Phylogenetic tree analysis indicated that AcIAA9 exhibited high homology to IAA9 from Camellia sinensis,and AcIAA9 belonged to the same branch in evolutionary tree as VvIAA9 from Vitis vinifera.Promoter analysis revealed that the sequence of AcIAA9 contained a variety of cis-elements,in addition to the basic elements that regulate eukaryotic gene transcription,such as TATA box and CAAT box,it also had a variety of stress response factors,such as anaerobic stress and drought stress.Real-time qPCR analysis showed that AcIAA9 had differential expression in tissues,and the expression was highest in flower.
