新疆石河子一株捕食线虫性真菌-圆锥节丛孢菌的分离鉴定与捕食线虫活性分析

摘  要:从新疆石河子地区天山北坡牧场土壤中分离培养捕食线虫性真菌,筛选高捕食线虫活性的菌株,为草食动物线虫病生防制剂研发奠定基础。采用土壤散布法、线虫诱饵法分离纯化捕食线虫性真菌,根据菌丝、分生孢子、捕食结构的形态特征及分子生物学方法对分离的菌株进行种属鉴定,并分析其遗传进化关系及捕食线虫活性。结果分离得到一株捕食线虫性真菌(命名为NTF-SHZ6菌株),该菌捕食结构为粘性菌网,可产生大量的分生孢子和少量的厚垣孢子。其中,分生孢子无色,呈倒圆锥,中有一隔,与节丛孢形态相符。分离株ITS1基因序列与圆锥节丛孢同源性为99%~100%,确定该菌株为圆锥节丛孢菌(Arthrobotrysco-noides)。捕食线虫活性测定显示,该菌对秀丽隐杆线虫的捕食率高达99.25%。A nematode-trapping fungus was isolated and cultured from the soil in pastures on the northern slope of the Tianshan Mountains in the Shihezi area of Xinjiang,and strains with high nematode-predatory activity were selected.This study provides a basis for developing biocontrol agents against nematode diseases in herbivores.The fungus was isolated and purified using soil dispersion and nematode baiting methods.The strains were identified at the species level based on the morphological characteristics of their mycelia,conidia,and trapping structures,as well as by molecular biological methods,to analyze their phylogenetic relationships and nematode-predatory activity.A nematodetrapping fungus(designated as NTF-SHZ6)was isolated,characterized by a sticky mycelial net that produces a large number of conidia and a few chlamydospores.The conidia are colorless,conical and have a septum,consistent with the morphology.The internal transcribed spacer 1(ITS1)gene sequence of the isolated strain showed 99%to 100%homology with Arthrobotrys conoides,confirming its identification as this species.The nematode-predatory activity assay revealed that this fungus exhibited a predation efficiency of 99.25%against Caenorhabditis elegans.
