摘 要:黄瓜(Cucumis sativus L.)是重要的蔬菜作物,也是研究单性花发育的模式植物。黄瓜雌花比例受到植物激素、温度和光周期等条件影响。前期研究发现乙烯受体基因CsETR1在雌花雄蕊发育过程中表达特异下调,参与雌花雄蕊的滞育过程。为研究CsETR1表达如何被调控,利用酵母单杂交试验筛选与CsETR1启动子相互作用的蛋白,发现其中包括表达受光周期调控的转录因子CsSTO(SALT TOLERANCE)。酵母双杂交和BiFC(双分子荧光互补)试验表明,CsSTO与花器官身份决定B类蛋白CsAP3相互作用。对CsSTO功能进行了初步探究发现,与拟南芥中STO基因类似,CsSTO表达也受到光诱导。利用雌激素诱导型载体在拟南芥中诱导表达CsSTO后,CsETR1启动子驱动的GUS报告基因表达上调,表明CsSTO在转基因拟南芥中可以激活CsETR1启动子表达。这些结果表明CsSTO很可能与CsAP3相互作用,整合光周期信号共同调控CsETR1转录,进而调控黄瓜花性别分化过程。Cucumber(Cucumis sativus L.)is an important vegetable and the model system for studying unisexual flower development.The proportion of female flowers in cucumbers is affected by phytohormones,temperature,and photoperiod.Previous study showed that the expression of the ethylene receptor gene CsETR1 is specifically down-regulated at female flower stamen primordium and involved in the arrest of female flower stamen.To investigate the underlying mechanism CsETR1 transcription regulation,this study utilized yeast one-hybrid assay to screen for proteins interacting with the CsETR1 promoter.The analysis revealed that a photoperiod-regulated transcription factor CsSTO(SALT TOLERANCE)in the promoter region.Yeast two-hybrid and BiFC(bimolecular fluorescence complementation)experiments demonstrated that CsSTO interacts with the floral organ identity-determining class B protein CsAP3.Similar to STO in Arabidopsis,CsSTO expression was light-induced.The expression of GUS reporter gene driven by CsETR1 promoter was upregulated when the expression of CsSTO was induced in transgenic Arabidopsis,which indicated that CsSTO can activate CsETR1 promoter expression.These results suggest that CsSTO is likely to interact with CsAP3 and integrate photoperiodic signals to co-regulate CsETR1 transcription,which,in turn,regulates the process of cucumber flower unisexual flower development.
