妇康栓对大鼠胚胎-胎仔发育毒性

摘　　要：目的 探讨妇康栓对大鼠胚胎-胎仔发育毒性。方法 SPF级SD大鼠[雄鼠30只、体重为(300±20)g,雌鼠60只、体重为(240±20)g],11周龄,按雄、雌1∶2合笼,将交配成功的40只孕鼠按照随机数字表法分为对照组和妇康栓低、中、高剂量组,每组10只。在妊娠第7~16天时,妇康栓低、中、高剂量组分别给予1.89、3.78、7.56 g/kg妇康栓,对照组给予等量的硬脂酸聚烃氧(40)酯。比较各组孕鼠的行为活动、毛发、呼吸、眼口鼻分泌物及死亡等一般状况;比较各组孕鼠体重、摄食量;比较各组黄体数,胎鼠存活率、身长、尾长、胎重、胎盘重;比较各组胎鼠的外观、内脏、骨骼情况。结果 四组孕鼠行为活动正常、毛发光泽自然、呼吸平稳、眼口鼻无异常分泌物,且未出现死亡等异常情况。四组孕鼠的体重和摄食量均稳定增长。四组胎鼠生存情况及黄体数比较,差异无统计学意义(P>0.05)。四组胎鼠身长、尾长、胎重、胎盘重比较,差异无统计学意义(P>0.05)。四组胎鼠头颈、躯干、四肢均未见异常,且皮下未见出血。四组胎鼠的头部、胸部、腹部内脏均未出现异常。四组胎鼠的指骨、趾骨、肋骨、胸骨等骨骼结构均发育良好,未出现任何畸形。结论 妇康栓对大鼠胚胎-胎仔无潜在的发育毒性和致畸性。Objective To observe the development toxicity of Fukang Suppository on rats embryo-fetal.Methods SPF SD rats[30 males with body weight of(300±20)g and 60 females with body weight of(240±20)g],aged 11 weeks,were caged 1∶2 males and females.A total of 40 pregnant rats were divided into control group and Fukang Suppository low,medium,and high dose groups,with 10 pregnant rats in each group according to random number table method.On the 7th to 16th day of gestation,Fukang Suppository low,medium,and high dose groups were given 1.89,3.78,7.56 g/kg,respectively,and control group was given the same amount of polyalkoxy stearate(40)ester.The behavior,hair,respiration,eye,mouth and nose secretions,and death of pregnant rats among four groups were compared;the weight and food intake of pregnant rats among four groups were compared;luteal number,and survival rate,body length,tail length,fetal weight,and placental weight of fetal rats among four groups were compared;and the appearance,viscera,and skeleton of fetal rats among four groups were compared.Results The pregnant rats of four groups had normal behavior,natural hair luster,stable breathing,no abnormal secretions from eyes,mouth and nose,and no abnormal conditions such as death.The weight and food intake of pregnant rats among four groups increased steadily.There were no significant differences in survival condition of fetal rats and luteal number among four groups(P>0.05).There were no significant differences in body length,tail length,fetal weight,and placental weight of fetal rats among four groups(P>0.05).No abnormality was found in the head and neck,trunk,and limbs of the four groups,and no subcutaneous bleeding of fetal rats was observed.There were no abnormalities in the head,chest,and abdominal organs of fetal rats among four groups.Phalanges,phalanx,ribs,sternum,and other skeletal structures of fetal rats among four groups were well developed without any deformity.Conclusion Fukang Suppository has no potential developmental toxicities and teratogenicities o
