加减复元活血汤调控TGF-β1/Smads通路抑制大鼠术后肠粘连实验研究

摘  要:目的 研究加减复元活血汤对大鼠术后肠粘连的防治作用并探讨其作用机制。方法 选取6周龄SPF级雄性SD大鼠35只,按照随机数字表法将其分为空白组、模型组、低剂量组(生药含量2.2 g/g)、中剂量组(生药含量4.4 g/g)、高剂量组(生药含量8.8 g/g),每组7只。空白组大鼠仅进行开腹、关腹处理,其余各组大鼠进行盲肠摩擦+对应腹壁损伤建立术后肠粘连模型。各组给药体积均为20 ml/kg,1次/d,连续给药7 d。实验结束后评估各组大鼠肠粘连情况,苏木精-伊红(HE)染色观察肠粘连处组织病理形态,RT-q PCR法检测肠粘连组织中转化生长因子-β1(TGF-β1)、Smad2、Smad3、Smad7及基质金属蛋白酶9(MMP-9)的m RNA表达水平,Western blot法检测肠粘连组织中TGF-β1、Smad2、Smad3、Smad7蛋白的表达水平,测定肠粘连组织中羟脯氨酸含量。结果 空白组大鼠均未见明显肠粘连,模型组大鼠见不同程度的肠粘连形成,病理学观察见明显炎症反应及肉芽组织增生。与模型组比较,低、中、高剂量组大鼠肠粘连程度降低(P<0.05)。与空白组比较,模型组TGF-β1、Smad2、Smad3m RNA及蛋白表达水平升高(P<0.05),Smad7蛋白表达水平降低(P<0.05),羟脯氨酸含量升高(P<0.05)。与模型组比较,低、中、高剂量组TGF-β1、Smad3 m RNA表达水平降低(P<0.01),低、中剂量组Smad2 m RNA表达水平降低(P<0.05或P<0.01)。与模型组比较,低、中、高剂量组TGF-β1、Smad2、Smad3蛋白表达水平降低(P<0.05或P<0.01),Smad7蛋白表达水平升高(P<0.05)。与模型组比较,低、中、高剂量组羟脯氨酸含量降低(P<0.05)。与低剂量组比较,中剂量组羟脯氨酸含量降低(P<0.05);与中剂量组比较,高剂量组羟脯氨酸含量升高(P<0.05)。低、中、高剂量组肠粘连程度,TGF-β1 m RNA及蛋白表达水平、Smad2 m RNA及蛋白表达水平、Smad3m RNA及蛋白表达水平、Smad7 m RNA及蛋白表达水平、MMP-9 m RNA表达水平比较,差异无Objective To study the preventive and therapeutic effects of Modified Fuyuan Huoxue Decoction on postoperative intestinal adhesion in rats and explore its mechanism.Methods Thirty-five SPF male SD rats at six weeks were selected.According to the random number table method,they were divided into five groups:blank group,model group,low dose group(crude drug 2.2 g/g),middle dose group(crude drug 4.4 g/g),and high dose group(crude drug 8.8 g/g),with seven rats in each group.in the blank group rats were only open and closed,and the remaining groups rats were performed on blind sausage friction+corresponding abdominal wall damage to establish a postoperative intestinal adhesion model.The dosage volume for each group was 20 ml/kg.Once a day,continuous administration for one week.After the experiment,the intestinal adhesion of each group was evaluated.Hema-toxylin-eosin(HE)staining was used to observe the histopa-thological morphology of intestinal adhesions. RT-qPCR was used to detect the mRNA expression levels of transforming growth factor -β1 (TGF-β1), Smad2, Smad3, Smad7, and matrix metalloproteinase 9 (MMP-9) in intestinal adhesion tissues. Western blot analysis was performed to detect the expression levels of TGF-β1, Smad2, Smad3, and Smad7 proteins in intestinal adhesion tissues, and to determine the content of hydroxyproline in intestinal adhesion tissues. Results No obvious intestinal adhesion was found in the blank control group, and the intestinal wall of the model control group was adhered to different degrees. Pathological observation showed obvious inflammatory reaction and granulation tissue hyperplasia. Compared with the model group, the degree of intestinal adhesion in each dose group was reduced (P<0.05). Compared with the blank group, the model group TGF-β1, Smad2, Smad3 mRNA and protein expression levels increased (P<0.05), the expression level of Smad7 protein decreased (P<0.05), and the content of hydroxyticine increased (P<0.05).
