摘 要:目的 探讨调脂通脉方调控TLR4/NF-κB信号通路改善缺血性脑卒中大鼠神经功能的作用机制。方法 选取8周龄SPF级雄性SD大鼠120只,体重(220±20)g,按照随机数字表法分为假手术组,模型组,调脂通脉方低、中、高剂量组及尼莫地平组,每组20只。除假手术组外,其余各组大鼠采用中脑动脉栓塞法造模,2 h后实现缺血再灌注。假手术组和模型组灌胃等量生理盐水,调脂通脉方低、中、高剂量组灌胃调脂通脉方20、40、60 g/(kg·d),尼莫地平组灌胃尼莫地平15 mg/(kg·d),共14 d。灌胃结束后对大鼠进行神经功能评分;比较各组脑梗死面积占比,超敏C反应蛋白(hs-CRP)、脂蛋白相关磷脂酶A2(Lp-PLA2)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量及TLR4、NF-κB表达水平。结果 与假手术组比较,模型组大鼠神经功能评分升高(P<0.05);与模型组比较,调脂通脉方低、中、高剂量组神经功能评分降低(P<0.05)。与假手术组比较,模型组大鼠脑梗死面积占比增加(P<0.05);与模型组比较,调脂通脉方低、中、高剂量组大鼠脑梗死面积占比降低(P<0.05)。与假手术组比较,模型组hs-CRP、Lp-PLA2、IL-6、TNF-α含量升高(P<0.05);与模型组比较,调脂通脉方低、中、高剂量组hs-CRP、Lp-PLA2、IL-6、TNF-α含量降低(P<0.05)。与假手术组比较,模型组TLR4、NF-κB蛋白表达水平升高(P<0.05);与模型组比较,调脂通脉方低、中、高剂量组TLR4、NF-κB蛋白表达水平降低(P<0.05)。结论 调脂通脉方可通过抑制TLR4/NF-κB通路,减少炎症因子释放,从而改善脑缺血再灌注损伤大鼠的神经功能。Objective To investigate the mechanism of regulating TLR4/NF-κB signaling pathway by Tiaozhi Tongmai Decoction to improve nerve function in ischemic stroke rats.Methods A total of 1208-week-old SPF male SD rats with body weight of(220±20)g were selected,and they were divided into sham operation group,model group,low-,medium-,and high-dose groups and Nimodipine group according to random number table method,with 20 rats in each group.Except sham operation group,the other groups of rats were modelled by middle cerebral artery occlusion and achieved ischemia-reperfusion after two hours.Sham operation group and model group were administrated with equal amount of normal saline,Tiaozhi Tongmai Decoction low-,medium-,and high-dose groups were administrated with Tiaozhi Tongmai Decoction 20,40,and 60 g/(kg·d),and Nimodipine group was administrated with Nimodipine 15 mg/(kg·d)for 14 days.The neural function of rats was evaluated after gavage;the proportion of cerebral infarction area,the contents of hypersensitive C-reactive protein(hs-CRP),lipoprotein-associated phospholipase A2 (Lp-PLA2), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and the expression levels of TLR4 and NF-κB in all groups were compared. Results Compared with sham operation group, neural function scores of rats in model group were increased (P<0.05);compared with model group, the neural function scores of Tiaozhi Tongmai Decoction low-, medium-, and high-dose groups were decreased (P<0.05). Compared with sham operation group, the proportion of cerebral infarction area in model group was increased (P<0.05);compared with model group, the proportion of cerebral infarction area in Tiaozhi Tongmai Decoction low-, medium-, and high-dose groups were decreased (P<0.05). Compared with sham operation group, the contents of hs-CRP, Lp-PLA2, IL-6, and TNF-α in model group were increased (P<0.05);compared with model group, the contents of hs-CRP, Lp-PLA2, IL-6, and TNF-α in Tiaozhi Tongmai Decoction low-, medium-, and high-dose groups were decrease
