摘　　要：目的分析河南地区人类白细胞抗原(HLA)、人类血小板抗原(HPA)基因供者库的库容情况及匹配概率。方法以1445名河南地区多次血小板捐献者为对象,采用多重荧光PCR方法对其进行HPA-1~6、-10、-15、-21和HLA-A、-B基因分型检测分析,应用Arlequin 3.5.2.2软件计算基因频率、单体型频率和表型频率,评估库容水平。结果在1445人份已知HPA、HLA基因分型结果的供者库中,计算推导找到1名HPA基因型相同供者的概率为91%;找到1名HLA-A、-B单体型相匹配供者概率为66%;找到1名HLA表型相匹配供者的概率为25%。结论建立了河南地区人群的HLA、HPA基因供者库,分析了河南地区HLA、HPA基因分布,为本地区血小板HLA、HPA基因供者库的建设、维护以及应用提供数据支持。Objective To analyze the capacity and matching probability of platelet human leukocyte antigen(HLA)and human platelet antigen(HPA)gene donor banks in Henan region.Methods A total of 1445 platelet donors from Henan province were selected as the study subjects.HPA-1-6,-10,-15,-21 and HLA-A,-B gene typing were performed using multiple fluorescence PCR method.The gene frequency,haplotype frequency and phenotype frequency of HLA/HPA was performed with Arlequin 3.5.2.2 software.The storage capacity level was evaluated.Results Among 1445 platelet donors with known HLA and HPA genotype,the probability of finding one donor with full-matched HPA genotypes was calculated to be 91%.The probability of finding one donor with the matching HLA-A,-B haplotype was 66%.The probability of finding a donor with matching HLA phenotype was 25%.Conclusion The HLA and HPA gene donor database has been established in Henan,and the distribution of HLA and HPA genes in Henan province has been analyzed.Providing the data support for the construction,maintenance and application of platelet HLA and HPA gene donor banks in the region.
