摘 要:目的评价不同加工方法山柰饮片对功能性消化不良(functional dyspepsia,FD)模型大鼠的保护作用。方法分别制备山柰趁鲜加工饮片及山柰传统晒干饮片后,采用碘乙酰胺联合小平台站立法建立FD大鼠模型,并随机分成空白组、模型组、阳性药组、山柰传统晒干低、高剂量组、山柰趁鲜加工低、高剂量组。测定各组大鼠拉尾排便阳性率及胃排空率;测定大鼠血清中淀粉酶(AMS)、D-木糖(D-xylose)、乳酸(LA)和胃动素(MOT)含量;HE染色观察胃组织病理情况;通过分子对接预测山柰治疗FD的核心成分。结果与模型组比较,趁鲜加工山柰饮片及传统晒干饮片可降低拉尾排便阳性率,提高胃排空率,显著增加血清AMS、D-xylose、MOT水平(P<0.01),显著降低乳酸含量(P<0.01),对FD模型有治疗作用;与山柰传统晒干高剂量饮片相比,山柰趁鲜加工高剂量饮片能更好地提高FD模型大鼠的食量及饮水量,改善其体质量,显著降低拉尾排便阳性率(P<0.05),提高AMS、D-xylose含量(P<0.05)及降低LA含量(P<0.01);给药组大鼠的胃窦组织病理HE染色切片,均未发现明显器质性改变,无溃烂情况;分子对接结果预测山柰治疗FD核心成分为山柰酚、山柰素、(R)-4-甲氧基黄檀醌、3,4-dimethoxydalbergione和对甲氧基肉桂酸乙酯(EPMC)。结论山柰趁鲜加工饮片及山柰传统晒干饮片对FD模型大鼠有一定的治疗作用,山柰趁鲜加工饮片的作用更佳。Objective To evaluate the protective effects of different processing methods of K.galanga tablets on rats with functional dyspepsia(FD).Methods After the preparation of freshly processed and traditionally dried K.galanga tablets respectively,the rat model of FD was established by using iodoacetamide combined with small platform station legislation,and randomly divided into the blank group,model group,positive drug group,low and high dose groups of K.galanga traditionally dried,and low and high dose groups of K.galanga processed while fresh.The positive rates of tail pulling and gastric emptying were determined.The serum levels of amylase(AMS),D-xylose,lactic acid(LA)and gastric motility(MOT)were measured in rats.HE staining was used to observe the gastric histopathology.The core constituents of Kaempferia kaempferi for the treatment of FD were predicted by molecular docking.Results Compared with the model group,while-fresh processed K.galanga drinking tablets and K.galanga while-fresh processed drinking tablets had therapeutic effects on the FD model,which could reduce the positive rate of tail pulling and defecation,improve the rate of gastric emptying,increase the levels of serum AMS,D-xylose,and MOT(P<0.01),and decrease the content of lactic acid(P<0.01).Compared with traditional sun-dried K.galanga tablets,K.galanga processed while fresh tablets increased the food and water intake of FD rats,improved body mass,reduced the positive rate of tail pulling and defecation(P<0.05),increased the content of AMS and D-xylose(P<0.05)and reduced the content of LA(P<0.01).The histopathological HE staining of sinusoids of the rats did not show any obvious organismal changes,and there was no ulceration.The molecular docking results showed that the core components of K.galanga therapeutic FD were kaempferol,(R)-4-methoxyflavanquinone,3,4-Dimethoxydalbergione and ethyl p-methoxycinnamate(EPMC).Conclusion Freshly processed and traditionally dried K.galanga tablets have certain therapeutic effects on FD rats,and K.galanga is be
