摘  要:目的建立国公酒高效液相色谱(HPLC)指纹图谱,并结合化学模式识别对其质量进行分析评价。方法采用HPLC建立国公酒的指纹图谱,采用Agilent Zorbax SB-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相:乙腈(A)-0.1%甲酸水(B),梯度洗脱,流速:1.0 ml/min,检测波长:283 nm,柱温:35℃,进样量:10μl。运用“中药色谱指纹图谱相似度评价系统(2012版)”对10批国公酒进行相似度评价,并通过液相色谱-质谱(LC-MS)对其共有峰进行鉴定。采用系统聚类分析(HCA)、主成分分析(PCA)与正交偏最小二乘判别分析(OPLS-DA)对10批国公酒的指纹图谱进行分析评价。结果建立了国公酒的HPLC指纹图谱,确定了33个共有峰,鉴定出16个化合物,分别为辛弗林、羟基红花色素A、升麻素苷、阿魏酸、芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、补骨脂素、异补骨脂素、丹皮酚、欧前胡素、羌活醇、和厚朴酚、蛇床子素、厚朴酚。10批样品相似度不低于0.945。PCA与OPLS-DA分析结果一致,可将10批样品分为2组,并筛选出3个差异性成分。结论本文建立的HPLC指纹图谱,可用于评价国公酒生产工艺的稳定性和产品质量的一致性,对国公酒的质量评价具有一定参考价值。Objective To establish the high performance liquid chromatography(HPLC) fingerprint of Guogong Jiu and evaluate its quality through combination with chemical pattern recognition.Methods The fingerprint of Guogong Jiu was established by HPLC.The analysis was performed on an Agilent Zorbax SB-C_(18) column(4.6mm×250 mm,5μm),using acetonitrile(A)-0.1 % formic acid water(B) as mobile phase of gradient elution.The flow rate was 1.0 ml/min,the detection wavelength was 283 nm,the column temperature was 35℃ and the injection volume was 10 μl.The similarity of 10 batches of Guogong Jiu was evaluated by Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(2012 Edition).The common peaks were identified by liquid chromatography-mass spectrometry(LC-MS).Hierarchical clustering analysis(HCA),principal component analysis(PCA),and orthogonal partial least-squares discrimination analysis(OPLS-DA) were used for the further assessment of 10 batches of Guogong Jiu samples.Results HPLC fingerprint of Guogong Jiu was established,33common peaks were obtained,and 16 compounds were identified,including synephrine,hydroxysafflor yellow A,prim-O-glucosylcimifugin,ferulic acid,narirutin,naringin,hesperidin,neohesperidin,psoralen,angelicin,paeonol,imperatorin,notopterol,honokiol,osthole and magnolol.The similarity of 10 samples was not less than 0.945.The results of PCA and OPLS-DA are consistent.10 batches of samples were classified into two clusters and 3 major chemical markers causing quality differences were screened out.Conclusion The fingerprint can be used to evaluate the stability of production process and the product quality consistency of Guogong Jiu.It has some reference value for the quality evaluation of Guogong Jiu.
