摘 要:目的为更好地评价槐米和槐花质量,建立一种同时测定槐米及槐花中芦丁、槲皮素、山柰酚、异鼠李素4种黄酮类成分的高效液相色谱(HPLC)分析方法。方法色谱柱为Agilent 5 TC-C_(18)(250 mm×4.6 mm,5μm),以乙腈(A)及0.2%醋酸水溶液(B)为流动相,柱温40℃,梯度洗脱;检测波长355 nm,流速为1 ml/min。结果4种黄酮成分分离效果良好,峰形尖锐。建立了4种黄酮的定量标准曲线。方法学考察结果表明,各黄酮保留时间与峰面积的RSD分别介于0.11%~0.40%与0.51%~1.14%之间,加标回收率介于95.42%~104.64%之间。结论芦丁是槐米和槐花中含量最高的黄酮成分,含量分别为131.57~181.10 mg/g,72.31~88.15 mg/g。芦丁、槲皮素和异鼠李素在槐米中含量显著高于槐花中含量,而山柰酚两者中含量差异不明显。Objective To establish a high-performance liquid chromatography(HPLC) method for the simultaneous determination of four flavonoids,namely rutin,quercetin,kaempferol and isorhamnetin in Flos Sophorae Immaturus and Sophorae Flos.Methods An Agilent 5 TC-C18 column(250 mm×4.6 mm,5 μm) was employed,using acetonitrile(A) and 0.2 % acetic acid solution(B) as mobile phase for gradient elution.The column temperature was 40℃,the detection wavelength was set at 355 nm,and the flow rate was 1 ml/min.Results The separation efficiency of the four flavonoids was excellent and the peak shape was sharp.The quantitative standard curves of four flavonoids were well established.Methodological investigation revealed that the RSDs for retention time and peak area of each flavonoid ranged from 0.11 % to 0.40 % and 0.51 % to 1.14 %,respectively,while the recoveries ranged from 95.42 % to 104.64 %.Conclusion Rutin was the predominant flavonoid component in Flos Sophorae Immaturus and Sophorae Flos,with levels ranging from 131.57 to 181.10 mg/g and 72.31 to 88.15 mg/g,respectively.The contents of rutin,quercetin and isorhamnetin in Flos Sophorae Immaturus were significantly higher than those in Sophorae Flos,while the content difference of kaempferol in Flos Sophorae Immaturus and Sophorae Flos was not significant.
