摘 要:目的基于传统煎药工艺,优化生脉饮(党参方)的制备工艺。方法按照传统煎药工艺,以8种指标成分含量和出膏率为指标,考察不同溶剂用量、浸泡时间和煎煮时间对提取工艺的影响以及不同浸膏浓缩程度和含醇量对纯化工艺的影响。结果生脉饮(党参方)最优制备工艺为精密称取党参15 g,麦冬10 g,五味子5 g,加入10倍量纯化水,浸泡30 min,电热套武火煮沸,换文火回流30 min,双层纱布趁热过滤,残渣加8倍量水,继续文火回流30 min,过滤,合并2次滤液,减压浓缩至0.8 g/ml,边搅拌边缓慢加入1倍量95%乙醇,静置过夜,3500 r/min离心10 min,取上清液旋干,加水定容到50 ml,得到生脉饮溶液。结论本实验优化的制备工艺合理可行,可为生脉饮相关制剂的开发和工业化生产提供一定的研究基础。Objective To optimize the preparation process of Shengmai Yin(Dangshen Formula)based ontraditional decoction techniques.Methods Following traditional decoction methods,the effects of different solventvolumes,soaking times,and decoction durations on the extraction process were investigated using the contents ofeight marker compounds and the extract yield as evaluation indicators.Additionally,the influence of different con-centrate densities and ethanol concentrations on the purification process was examined.Results The optimalpreparation process for Shengmai Yin(Dangshen Formula)was as follows:precisely weigh 15 g of Codonopsis Pi-losula,10 g of Ophiopogon,and 5 g of Schisandra chinensis,add 10 times the amount of purified water,soak for30 min,bring to a vigorous boil using an electric heating mantle,then simmer under reflux for 30 min.Filter thehot solution through double-layered gauze,add 8 times the amount of water to the residue,and continue simmeringunder reflux for another 30 min.Combine the two filtrates and concentrate under reduced pressure to a density of0.8 g/ml.Slowly add an equal volume of 95%ethanol while stirring,let stand overnight,centrifuge at 3500 rpmfor 10 min,collect the supernatant,evaporate to dryness,and dilute with water to a final volume of 50 ml to obtainthe Shengmai Yin solution.Conclusion The optimized preparation process developed in this study is reasonableand feasible,providing a research foundation for the development and industrial production of Shengmai Yin-relat-ed formulations.
