再造生血胶囊治疗化疗后骨髓抑制的效应和机制研究

摘  要:目的:研究再造生血胶囊对化疗诱导的骨髓抑制的治疗作用及机制。方法:将雄性BALB/c小鼠随机分为正常组、模型组、再造生血胶囊治疗组,每组15只。除正常组外,其余各组均采用腹腔注射环磷酰胺(100 mg/kg)建立骨髓抑制小鼠模型。造模成功后,治疗组经口灌胃给予再生造血胶囊(624 mg/kg),3次/d,连续21 d。治疗结束后,检测小鼠外周血象变化,采用吉姆萨染色观察骨髓细胞形态和双抗体ABC-ELISA法检测各组造血生长相关因子的含量。结果:与对照组相比,模型组小鼠白细胞计数、红细胞计数、血红蛋白及血小板计数、骨髓有核细胞明显减少,骨髓细胞密度显著降低(P<0.05)。与模型组相比,治疗组小鼠在连续用药第21天后的白细胞计数、血红蛋白水平及血小板计数显著增加(P<0.05,P<0.01)、在连续用药第14、21天时红细胞计数均显著上升(P<0.05、P<0.01)。与模型组相比,治疗组小鼠的骨髓有核细胞数量增加(P<0.01),骨髓细胞密度有所改善,造血细胞丰富,骨髓中人促血小板生成素(Thrombopoietin,TPO)、促红细胞生成素(erythropoietin,EPO)水平显著上升(P<0.05,P<0.01)。结论:再造生血胶囊具有升高外周血红细胞、白细胞、血红蛋白及血小板计数,改善骨髓抑制的作用,其机制可能与增加骨髓中TPO、EPO水平有关。Objective:To study the therapeutic effect and mechanism of Zaizao Shengxue capsule on chemotherapy-induced myelosuppression.Methods:Male BALB/c mice were randomly divided into normal control group,model group and Zaizao Sheng Xue capsule treatment group,with 15 mice in each group.Except the normal control group,all the other groups were treated with intraperitoneal injection of cyclophosphamide(100 mg/kg)to establish myelopyrosuppressive mouse model.After successful modeling,the treatment group was given regenerated hematopoietic capsule(624 mg/kg)by oral gavage,3 times/day,for 21 consecutive days.After treatment,the peripheral blood changes were detected,and bone marrow cell morphology was observed by Giemsa staining and the content of hematopoietic growth-related factors was detected by double antibody ABC-ELISA.Results:Compared with the control group,mice in the model group showed a significant decrease in white blood cell count,red blood cell count,hemoglobin,and platelet count,as well as a marked reduction in bone marrow nucleated cell count and bone marrow cell density(P<0.05).Compared with the model group,mice in the treatment group exhibited a significant increase in white blood cell count,hemoglobin levels,and platelet count after 21consecutive days of drug administration(P<0.05,P<0.01),and a significant rise in red blood cell count on the 14th and 21st days of continuous drug administration(P<0.05,P<0.01).Compared with the model group,mice in the treatment group had an increased number of bone marrow nucleated cells(P<0.01),improved bone marrow cell density,enriched hematopoietic cells,and a significant increase in the levels of human thrombopoietin(TPO)and erythropoietin(EPO)in the bone marrow(P<0.05,P<0.01).Conclusion:Zaishengxue capsules have the effect of increasing the peripheral blood red blood cell,white blood cell,hemoglobin,and platelet counts,and improving bone marrow suppression.The mechanism may be related to the increased levels of TPO and EPO in the bone marrow.
