摘  要:根系分泌物影响根际微生物群落组成,是引起植物连作生物障碍的重要驱动因子。利用气相色谱质谱联用技术(GC-TOF-MS)研究新植健康及连作发病香草兰根系分泌物组成特征,共检测到83种代谢产物,包括脂肪酸、糖、氨基酸、酚酸、醇、胺、酯、苷、吡啶、嘧啶、醛、尿素等;主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)结果表明,新植健康及连作发病香草兰根系分泌物分别聚集在一起,可明显区分开来;运用OPLS-DA、模型的变量进行重要性因子和方差分析发现,尿素、乳糖、甘油二酯、柠檬酸、D-甘油-1-磷酸、4-羟基丁酸钠是差异代谢物,新植健康香草兰根系乳糖相对含量显著低于连作发病香草兰,而其他5种差异代谢物相对含量显著高于连作发病香草兰。平板对峙试验结果表明,添加不同浓度的柠檬酸均对香草兰枯萎病病原菌尖孢镰刀菌(Fusarium oxysporum sp.vanillae)具有抑制作用,而添加200 mmol/L的乳糖对尖孢镰刀菌生长具有先促进后抑制的作用。Root exudates have a vital effect on the rhizosphere microbial community structure and composition.They are key driving factors for plant continuous cropping barriers.In the present research,gas chromatography-mass spectrometry(GC-TOF-MS)was used to evaluate the composition characteristics of root exudates secreted by newly planted healthy and continuously cropped vanilla orchids.Eighty-three metabolites were detected,including fatty acids,sugars,amino acids,phe-nolic acids,alcohols,amines,esters,glycosides,pyridines,pyrimidines,aldehydes,urea,etc.,were detected.Principal com-ponent analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)revealed that the root exudates of newly planted healthy and continuously cropped diseased vanilla orchids were clustered alongside and could be clearly dis-tinguished.Urea,lactose,diglyceride,citric acid,D-glycerol-1-phosphate,and sodium 4-hydroxybutyrate were the differen-tially abundant metabolites.The relative content of lactose in the roots of newly planted healthy vanilla orchids was signifi-cantly lower than that in the roots of continuously cropped diseased vanilla orchids.However,the relative contents of the other five differentially abundant metabolites were remarkably greater than those in the roots of continuously cropped diseased va-nilla orchids.The antagonistic tests revealed that increasing different concentrations of citric acid significantly inhibited Fusarium oxysporum sp.vanillae(Fov).However,an additional 200 mmol·L-1 lactose promoted Fov growth,followed by inhibition.
