摘 要:试验选取24头胎次相近的初生仔猪(Landrace×Large×Yorkshire),根据7日龄体重,随机分为3组,每组8个重复,每个重复1头仔猪,分别为健康对照组(control)、出生后生长迟缓仔猪组(PGR)以及出生后生长迟缓仔猪+25 mg/kg BWβ-羟基丁酸组(PBHB)。试验期为49 d。研究结果表明:①与对照组相比,PGR组仔猪表现出明显的生长抑制;与PGR组相比,灌胃β-羟基丁酸显著增加了PBHB组的终末体重、平均日增重及股骨骨长(P<0.05)。②与对照组相比,PGR组仔猪回肠的绒毛高度(VH)、绒隐比(VCR)以及杯状细胞数量显著下降(P<0.05),而隐窝深度(CD)显著升高(P<0.05);与PGR组相比,PBHB组显著提高了VH、VCR以及杯状细胞数量(P<0.05),CD则显著下降(P<0.05)。③与对照组相比,PGR组回肠组织中干细胞增殖与分化相关基因Lgr5、Olfm4、Hes1、Notch1的相对表达量显著上升(P<0.05),Bmi1、ChgA、Muc2、Atoh1、Dll1、Notch1基因的相对表达量显著下降(P<0.05);PBHB组仔猪回肠组织中Lgr5、Olfm4、Bmi1、Hes1和Notch1基因相对表达量显著低于PGR组(P<0.05),而ChgA、Muc2、Atoh1、Dll1基因的相对表达量显著高于PGR组(P<0.05);同时,与PGR组相比,PBHB组仔猪回肠中肠内分泌细胞数量、ChgA蛋白丰度显著升高(P<0.05),Lgr5蛋白丰度有降低的趋势。④添加不同浓度BHB体外培养猪肠道类器官,与对照组相比,0.1 mmol/L和1 mmol/L BHB处理后出芽类器官数量及出芽效率显著升高(P<0.05)。0.1 mmol/L和1.0 mmol/L BHB处理组肠道类器官中Muc2和Dll1的基因相对表达量显著高于对照组(P<0.05),而Lgr5、Hes1的基因相对表达量显著低于对照组(P<0.05)。与对照组相比,1.0 mmol/L BHB处理显著降低了肠道类器官Olfm4的基因相对表达量(P<0.05)。此外,随着BHB浓度的增加,肠道类器官ChgA的基因相对表达量呈剂量依赖性增加(P<0.05)。综上所述,BHB能够提升出生后生长发育迟缓仔猪的生长性能,可能通过抑制Notch信号通路促进肠This study aimed to explore the effects of BHB on growth and intestinal development in post-natal growth-restricted piglets.A total of 24 newborn piglets(Landrace×Large×Yorkshire)with similar parity were selected and randomly divided into three groups based on their body weight at 7 days of age,with 8 replicates per group and 1 piglet per replicate.The groups were as follows:healthy controls(control group),postnatal growth retardation group(PGR),and postnatal growth retardation with 25 mg/kg BW BHB group(PBHB).After 49 days,results showed:①compared to the control group,PGR piglets exhibited growth suppression,while BHB significantly increased final body weight,average daily gain,and femur length in PBHB(P<0.05).②Compared with the control group,villus height(VH),pillow-to-crypt ratio(VCR),and goblet cell number in the ileum of piglets in the PGR group were significantly decreased,while crypt depth(CD)was significantly increased(P<0.05).In PGR piglets,BHB significantly increased the number of VH,VCR and goblet cells,while CD significantly decreased(P<0.05).③Compared to the control group,the relative expression levels of genes related to stem cell proliferation and differentiation(Lgr5,Olfm4,Hes1,Notch1)in the ileum tissue of the PGR group were significantly increased(P<0.05),while the relative expression levels of Bmi1,ChgA,Muc2,Atoh1,Dll1,and Notch1 genes were significantly decreased(P<0.05).PBHB normalized expression levels of key genes like ChgA,Muc2,Atoh1,and Dll1(P<0.05).Additionally,compared to the PGR group,the number of enteroendocrine cells and the protein abundance of ChgA in the ileum of the PBHB group were significantly increased(P<0.05),while the protein abundance of Lgr5 showed a decreasing trend.④In intestinal organoids,0.1 mmol/L and 1.0 mmol/L BHB treatment significantly increased the number and germination efficiency of budding organoids,as well as the relative expressions of Muc2 and Dll1 genes,but decreased the relative expressions of Lgr5 and Hes1 genes(P<0.05).Treatment with 1.0 mmol
