摘 要:目的基于UHPLC-Q-Exactive Orbitrap HRMS技术鉴定仙鹤草中的化学成分。方法色谱分离采用ACQUITY UPLC BEH C_(18)色谱柱(100 mm×2.1 mm,1.7μm),以体积分数为0.1%甲酸水溶液(A)-甲醇(B)为流动相,梯度洗脱,流速为0.3 mL·min^(-1),柱温为40℃。质谱采用加热电喷雾离子源,在正负离子一级全扫描结合数据依赖二级扫描模式(Full MS/dd-MS^(2))下采集数据。运用Xcalibur 4.2软件,根据化合物保留时间、准分子离子峰和二级质谱信息,结合对照品及文献报道,对仙鹤草的化学成分进行解析。结果仙鹤草醇提液中共鉴定出130种化学成分,包括41种黄酮类、29种酚酸类、24种木脂素类、14种香豆素类、8种萜类、5种间苯三酚衍生物类、4种脂类及5种其他类成分。结论首次采用UHPLC-Q-Exactive Orbitrap HRMS对仙鹤草的化学成分进行了研究,该研究为仙鹤草的质量控制、药效物质基础研究以及综合开发应用提供了重要的理论依据。Objective To identify the chemical constituents of Agrimonia pilosa based on UHPLC-Q-Exactive Orbitrap HRMS technology.Methods The gradient elution was conducted on an ACQUITY UPLC BEH C_(18) column(100 mm×2.1 mm,1.7 μm) with a mobile phase of 0.1% formic acid aqueous solution(A)-methanol(B) at a flow rate of 0.3 mL·min~(-1) and a column temperature of 40 ℃.The mass spectra were acquired using a heated electrospray ionization source(H-ESI) in positive and negative ion primary full scan combined with data-dependent secondary scan mode(Full MS/dd-MS~2).The chemical constituents of Agrimonia pilosa were analyzed by Xcalibur 4.2 according to the retention time,excimer ion peak and secondary mass spectrum,in combination with the reference substance and literature reports.Results A total of 130 chemical components,including 41 flavonoids,29 phenolic acids,23 lignans,14 coumarins,8 terpenoids,5 phloroglucinol derivatives,4 lipids,and 6 other components,were identified in the alcoholic extract of Agrimonia pilosa.Conclusion The chemical components of Agrimonia pilosa have been identified by UHPLC-Q-Exactive Orbitrap HRMS for the first time,which provides the theoretical basis for quality control,pharmacodynamic material basis research,and comprehensive development and application of Agrimonia pilosa.
