摘 要:为了探究库页红景天黄酮组分的体外抗氧化、抗炎作用及其对类风湿性关节炎(RA)的治疗作用和分子机制,本试验利用前期已建立的提取纯化方法获得较高纯度的库页红景天黄酮组分,采用超氧阴离子测定法和铁离子还原/抗氧化能力(RAP)法测定其抗氧化能力。建立小鼠巨噬细胞(Raw264.7)体外炎症模型,通过检测不同浓度(1、2、4 mg/mL)库页红景天黄酮组分处理后细胞上清液中炎症因子含量,分析其体外抗炎作用;以氧自由基吸收能力(ORAC)检测法评价其抗氧化能力。利用完全弗氏佐剂建立大鼠佐剂型关节炎模型,灌胃不同剂量(2、4、6 mg/mL)的库页红景天黄酮组分后,通过检测大鼠踝关节周长、血液中炎症因子和骨代谢相关信号通路因子含量、踝关节骨代谢相关信号通路蛋白表达和组织学检查,研究库页红景天黄酮组分对RA的治疗作用和潜在分子机制。结果显示,经提取纯化后的库页红景天黄酮组分具有良好的超氧阴离子清除效果,可提高FRAP的抗氧化效果;不同浓度的库页红景天黄酮组分均可显著抑制Raw264.7细胞促炎因子肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)和前列腺素2(PEG2)的分泌(P<0.05或P<0.01),提高ORAC值(P<0.05或P<0.01)。动物试验结果显示,RA模型建立成功,与模型组相比,库页红景天黄酮组分中剂量组大鼠踝关节肿胀程度极显著降低(P<0.01),大鼠血清中炎症因子TNF-α、IL-1β和PEG2含量均显著降低(P<0.05或P<0.01)、核因子κB受体活化因子配体(RANKL)/骨保护素(OPG)值显著降低(P<0.05),踝关节组织中核因子-kappaB(NF-κB)和RANKL蛋白表达水平极显著降低(P<0.01)、OPG蛋白表达水平极显著升高(P<0.01)、RANKL/OPG值极显著降低(P<0.01),关节炎指数评分显著降低(P<0.05)。结果表明,库页红景天黄酮组分具有较好的体外抗氧化和抗炎作用,对RA大鼠有降低踝关节肿胀程度、降低血液中炎症因子释放�To investigate the in vitro antioxidant and anti-inflammatory effects,as well as the therapeutic potential and molecular mechanisms of Rhodiola sachalinensis flavonoid components(RSF)in rheumatoid arthritis(RA),this study utilized a previously established extraction and purification method to obtain high-purity RSF.The superoxide anion scavenging assay and ferric reducing antioxidant power(FRAP)assay were used to evaluate antioxidant capacity.An in vitro inflammation model was established using Raw264.7 murine macrophages.The anti-inflammatory effects of different RSF concentrations(1,2,and 4 mg/mL)were assessed by measuring the levels of inflammatory cytokines in the culture supernatant.Oxygen radical absorbance capacity(ORAC)assay was used to further assess antioxidant activity.An adjuvant-induced arthritis model was established in rats using complete Freund’s adjuvant,and RSF was administered orally at different doses(2,4,and 6 mg/mL).The therapeutic effects and potential mechanisms were investigated by measuring ankle circumference,serum inflammatory cytokine levels,bone metabolism-related signaling factors,protein expression in the ankle joint,and histopathological changes.The results showed that the purified RSF exhibited strong superoxide anion scavenging ability and significantly enhanced FRAP antioxidant effects.RSF at different concentrations significantly inhibited the secretion of pro-inflammatory cytokines,including tumor necrosis factor-alpha(TNF-α),interleukin-1β(IL-1β),and prostaglandin-2(PEG2)in Raw264.7 cells(P<0.05 or P<0.01),while increasing ORAC values(P<0.05 or P<0.01).In animal experiments,the RA model was successfully established.Compared with the Model group,the Medium-dose RSF group exhibited a significant reduction in ankle swelling(P<0.01),as well as significantly lower serum levels of TNF-α,IL-1β,and PEG2(P<0.05 or P<0.01).The receptor activator of nuclear factor-κB ligand(RANKL)/osteoprotegerin(OPG)ratio was significantly reduced(P<0.05).Furthermore,protein expression levels
