肾炎消白颗粒对糖尿病肾病模型大鼠足细胞synaptopodin表达影响的研究

摘　　要：目的:探究肾炎消白颗粒对糖尿病肾病模型大鼠足细胞synaptopodin表达的影响。方法:选择100只健康清洁级雄性SD大鼠,之后创建糖尿病肾病模型,创建的肾炎消白颗粒组、缬沙坦组、模型组和空白组,每组25只。2.5%胆固醇、10%猪油、20%蔗糖添加于常规饲料中,以高糖高脂饮食进行造模,成功造模后进行8周的药物干预,在2周、4周、6周、8周时分批将大鼠处死,测体重,并检测血肌酐、尿素氮、24 h尿蛋白定量,对肾组织进行HE染色,以免疫组化检测骨架蛋白synaptopodin表达。结果:空白组与模型组、缬沙坦组、肾炎消白颗粒组灌胃后体重、24 h尿蛋白定量、血清Scr、血BUN、synaptopodin蛋白灰度值对比,差异显著(P<0.05)。模型组灌胃后的体重均明显低于缬沙坦组、肾炎消白颗粒组,24 h尿蛋白定量、血清Scr、血BUN、synaptopodin蛋白灰度值高于缬沙坦组、肾炎消白颗粒组(P<0.05)。缬沙坦组、肾炎消白颗粒组灌胃后的体重、24 h尿蛋白定量、血清Scr、血BUN、synaptopodin蛋白灰度值无明显对比差异(P>0.05)。结论:肾炎消白颗粒可以降低糖尿病肾病大鼠尿蛋白定量,并将synaptopodin蛋白表达水平上调,减轻肾功能损伤,且肾炎消白颗粒获得的效果与西药缬沙坦较为相似。Objective:To explore the effect of Shenyan Xiaobai Granules on the expression of synaptopodin in podocytes of diabetic nephropathy model rats.Methods:Select 100 healthy clean-grade male SD rats,and then create a diabetic nephropathy model.The Shenyan Xiaobai Granule group,valsartan group,model group and blank group were created,with 25 rats in each group.2.5%cholesterol,10%lard,and 20%sucrose were added to the conventional feed,and the model was modeled with a high-sugar and high-fat diet.After successful modeling,8 weeks of drug intervention was performed.At 2 weeks,4 weeks,6 weeks,and 8 Rats were sacrificed in batches every week,and their body weights were measured.Serum creatinine,urea nitrogen,and 24 h urine protein were also measured.HE staining of kidney tissue was performed,and the expression of the skeleton protein synaptopodin was detected by immunohistochemistry.Results:The body weight,24 h urine protein quantification,serum SCr,blood BUN,and synaptopodin of the blank group and the model group,valsartan group,and Shenyan Xiaobai Granules group after intragastric administration Comparison of protein gray value showed significant difference(P<0.05).The body weight of the model group after intragastric administration were all significantly lower than those of the valsartan group and the Shenyan Xiaobai Granules group.The 24-hour urine protein quantification,serum Scr,blood BUN,and synaptopodin The protein gray value was higher than that of the valsartan group and the Shenyan Xiaobai Granules group(P<0.05).There was no significant difference in body weight,24 h urine protein quantification,serum Scr,blood BUN,and synaptopodin protein gray value in the valsartan group and the Shenyan Xiaobai Granule group after oral administration.Compare the differences(P>0.05).Conclusion:Shenyan Xiaobai Granules can reduce urinary protein quantification in rats with diabetic nephropathy,increase the expression level of synaptopodin protein,and reduce renal function damage.The effect obtained by Shenyan Xiaobai Granules is
