摘 要:目的:研究血府逐瘀汤对心房颤动(AF)模型大鼠的心电图改变以及炎症反应、氧化应激等指标的影响,深入探究血府逐瘀汤对AF大鼠的治疗作用及其影响机制。方法:60只雌性大鼠随机分成空白组,模型组,维拉帕米组,血府逐瘀汤高、中、低剂量组,每组10只。除空白组外,其余各造模组连续7 d尾静脉注射氯化钙(CaCl_(2))-乙酰胆碱(Ach)混合液建立房颤模型,7 d造模成功后,血府逐瘀汤高、中、低剂量组分别给予血府逐瘀汤汤剂14.04、7.02、3.51 g·kg^(-1)·d^(-1)灌胃,维拉帕米组给予维拉帕米混悬液25 mg·kg^(-1)·d^(-1)灌胃,空白组和模型组给予10 mL^(-1)·kg^(-1)·d^(-1)生理盐水灌胃,连续2周,同时仍进行尾静脉注射Ach-CaCl_(2)溶液。用生物信号采集器监测AF大鼠诱发及持续时间,酶联免疫吸附试验(ELISA)检测各组大鼠血清中肿瘤坏死因子α(TNF-α)、超敏C-反应蛋白(hs-CRP)、活性氧(ROS)、超氧化物歧化酶(SOD)、心肌肌钙蛋白T(cTnT)含量。采用HE染色法和Masson染色法观察各组大鼠左心房的病理改变。结果:模型组大鼠精神萎靡、爪甲青紫、眼球充血,经血府逐瘀汤干预后,一般情况明显好转。心电图结果显示,与模型组比较,血府逐瘀汤各剂量组和维拉帕米组显著延长AF诱发时间,缩短AF持续时间(P<0.01),以血府逐瘀汤高剂量组效果最佳。ELISA结果显示,与模型组比较,血府逐瘀汤各剂量组和维拉帕米组显著降低AF大鼠血清中hs-CRP、TNF-α、cTnT和ROS水平(P<0.01),增加SOD水平(P<0.01),以血府逐瘀汤高剂量组效果最佳。HE染色和Masson染色结果显示,与模型组比较,血府逐瘀汤各剂量组和维拉帕米组使AF大鼠心肌纤维排列相对整齐,组织间隙仅有少量胶原纤维沉积,以血府逐瘀汤高剂量组效果最佳。结论:血府逐瘀汤可以改善大鼠的一般情况,延长AF诱发时间,缩短AF持续时间,改善心肌损伤及心房纤维化程度,其机制可�Objective:To investigate the effects of Xuefu Zhuyu Decoction on electrocardiogram changes,inflammatory response,oxidative stress,and other indicators in atrial fibrillation(AF)model rats,and to explore its therapeutic effects and underlying mechanisms.Methods:Sixty female rats were randomly divided into a blank group,model group,verapamil group,and high-,medium-,and low-dose Xuefu Zhuyu Decoction groups,with 10 rats in each group.Except for the blank group,AF models were established in the other groups by continuous tail vein injection of a calcium chloride(CaCl_(2))-acetylcholine(Ach)mixed solution for 7 days.After successful modeling,the high-,medium-,and low-dose Xuefu Zhuyu Decoction groups were administered 14.04,7.02,and 3.51 g·kg^(-1)·d^(-1)of Xuefu Zhuyu Decoction,respectively,by gavage.The verapamil group was given 25 mg·kg^(-1)·d^(-1)of verapamil suspension,while the blank and model groups received 10 mL·kg^(-1)·d^(-1)of saline by gavage for 2 weeks,during which tail vein injection of the Ach-CaCl2 solution continued.A biological signal acquisition system was used to monitor the induction and duration of AF in rats.Enzyme-linked immunosorbent assay(ELISA)was employed to measure serum levels of tumor necrosis factor-α(TNF-α),high-sensitivity C-reactive protein(hs-CRP),reactive oxygen species(ROS),superoxide dismutase(SOD),and cardiac troponin T(cTnT).Hematoxylin-eosin(HE)staining and Masson staining were used to observe pathological changes in the left atrium of rats.Results:Rats in the model group exhibited lethargy,cyanosis,and conjunctival congestion,which significantly improved after Xuefu Zhuyu Decoction intervention.Electrocardiogram results showed that,compared with the model group,both the Xuefu Zhuyu Decoction and verapamil groups significantly prolonged AF induction time and shortened AF duration(P<0.01),with the high-dose Xuefu Zhuyu Decoction group showing the best effect.ELISA results indicated that,compared with the model group,the Xuefu Zhuyu Decoction and verapamil groups signifi
