摘  要:为深入探究引起毁灭性林木病害——黄栌枯萎病病原菌大丽轮枝菌Verticillium dahliae的致病机制,通过PEG介导原生质体遗传转化法获得黄栌枯萎病菌bZIP转录因子VdbZIP2突变体,开展VdbZIP2突变体的生物学性状分析,并进行致病性测定。结果表明:敲除VdbZIP2明显影响了黄栌枯萎病菌黑色素合成、微菌核发育、细胞壁胁迫敏感性以及对速效氮源的利用,VdbZIP2不是毒性必需的基因。明确黄栌枯萎病菌转录因子VdbZIP2参与微菌核形成、逆境胁迫和氮源利用过程,为后续深入解析VdbZIP2的分子调控网络提供重要线索。To deeply investigate the pathogenic mechanism of Verticillium dahliae,the causal agent of the devastating forest disease verticillium wilt of smoke tree,VdbZIP2,a bZIP transcription factor gene was deleted by PEG-mediated protoplast genetic transformation method.The biological traits of the mutants were analyzed,and the pathogenicity was determined.The results showed that knockout of VdbZIP2 seriously affected melanin synthesis,microsclerotia formation,cell wall stress sensitivity,and available nitrogen source utilization in V.dahliae.However,the mutants were fully pathogenic,indicating VdbZIP2 is not a virulence‐essential gene.In summary,we conclude that the transcription factor VdbZIP2 of V.dahliae is involved in microsclerotia formation,adversity stress and nitrogen source utilization,which provides important clues for the subsequent in‐depth analysis of the molecular regulatory network of VdbZIP2.
