摘  要:目的:本研究旨在通过高通量测序分析探究针刺足少阳经穴对去卵巢大鼠(OVX)骨质疏松的治疗效果,并阐述其对骨髓间充质干细胞调控骨平衡的潜在机制。方法:大鼠分为假手术组、模型组和电针组,并采集其股骨和胫骨样本,运用microCT、免疫组化及骨髓间充质干细胞高通量RNA测序技术进行分析。结果:电针显著抑制OVX诱导的腰椎和股骨骨质流失,提高了骨密度、骨小梁数、骨小梁厚度和骨小梁间距,降低了骨小梁分离度;电针能够在微观结构上明显改善骨转化相关指标的表达水平,促进OVX大鼠成骨标志物Runx2、Collagen I和Ocn的表达;电针能够促进治疗后骨髓间充质干细胞的成骨分化;对提取的股骨和胫骨的骨髓间充质干细胞进行高通量测序分析后发现,相关基因包括Csrp2、Dennd3以及Gldn等13个差异蛋白,涉及细胞增殖分化、免疫、血管新生和炎症等生物过程,最主要的信号通路包括细胞因子受体相互作用、破骨细胞分化、细胞粘附分子/细胞黏附分子以及趋化因子信号通路。结论:经电针治疗后去卵巢大鼠能够明显延缓骨质疏松的进展,同时还具有调控骨髓间充质干细胞成骨分化的能力。通过对相关基因的高通量RNA测序技术分析,笔者团队预测针刺足少阳经穴可能通过影响骨髓间充质细胞的成骨分化潜力,介导骨重塑平衡,改善骨质疏松的进展。Objective:To investigate the therapeutic effects of needling Foot Shaoyang meridian points on osteoporosis in ovariectomized(OVX)rats through high-throughput sequencing analysis and to explore the potential mechanisms by which it regulates bone homeostasis via bone marrow mesenchymal stem cells(BM-MSCs).Methods:45 SD rats were divided into the sham operation group,the model group,and the electro-acupuncture(EA)group.Femur and tibia samples were collected for analyses of Micro CT,immunohistochemistry and high-throughput RNA sequencing of BM-MSCs.Results:EA significantly inhibited OVX-induced bone loss in the lumbar spine and femur,improved bone mineral density,trabecular number,trabecular thickness and trabecular spacing,and reduced trabecular sepration.EA significantly improved the expression levels of bone remodeling-related indicators at the microstructural level and promoted the expression of osteogenic markers Runx2,Collagen I,and Ocn in OVX rats.EA promoted the osteogenic differentiation of BM-MSCs after the treatment.High-throughput sequencing analysis of BM-MSCs from femur and tibia samples revealed 13 differential proteins,including Csrp2,Dennd3,Gldn,involved in cellular proliferation,differentiation,immune response,angiogenesis and inflammation,and the major signaling pathways included cytokine receptor interaction,osteoclast differentiation,cell adhesion molecules/cell adhesion molecule and chemokine signaling pathways.Conclusion:After EA treatment,OVX rats exhibited a significant delay in the progression of osteoporosis,along with the ability to regulate the osteogenic differentiation of BM-MSCs.Through high-throughput RNA sequencing analysis of relevant genes,it is predicted that needling Foot Shaoyang points may mediate bone remodeling balance and improve the progression of osteoporosis by affecting the osteogenic differentiation potential of BM-MSCs.
