穴位电刺激对大鼠肝纤维化损伤的保护作用

摘  要:目的:观察穴位电刺激足三里、肝俞、三阴交、关元与太冲穴对大鼠肝纤维化损伤的保护作用,并探讨其机制。方法:将36只SPF级雄性SD大鼠随机分为空白组、模型组与穴位电刺激治疗组,每组12只。模型组、治疗组采用CCL4橄榄油溶液建立肝纤维化模型,治疗组于造模结束后1 d对足三里、肝俞、三阴交、关元和太冲穴进行电刺激治疗,每次干预15 min,1次/d,连续干预21 d。于造模结束及干预21 d后取材,对各组大鼠肝组织通过苏木素-伊红(HE)染色法和Massson染色法观察大鼠肝脏病理变化和肝纤维化程度,酶联免疫吸附试验(ELISA)法检测大鼠血清中透明质酸(HA)、层粘连蛋白(LN)和三型前胶原(PCⅢ)含量,实时荧光定量PCR法检测肝组织内α-SMA和Collagen1基因mRNA的相对表达水平。结果:通过对大鼠肝组织进行HE、Masson染色,模型组大鼠大量肝细胞出现坏死,且有胶原纤维堆积,治疗组胶原纤维堆积减少,肝细胞结构趋于正常;ELISA法检测,相较于空白组,模型组大鼠血清内HA、LN与PCⅢ含量明显上升,治疗组HA、LN与PCⅢ含量降低;实时荧光定量PCR实验验证模型组肝组织中肝纤维化指标α-SMA、Collagen1 mRNA含量升高,差异具有统计学意义(P<0.05),证明模型组造模成功,治疗组α-SMA、Collagen1 mRNA含量明显降低,差异具有统计学意义(P<0.05)。结论:穴位电刺激足三里、肝俞、三阴交、关元与太冲穴对肝纤维化损伤有一定的保护作用。Objective:To observe the protective effects of electro-needling Zusanli(ST36),Ganshu(BL18),Sanyinjiao(SP6),Guanyuan(RN4)and Taichong(LR3)on hepatic fibrosis injury in rats,and to explore its mechanisms.Methods:36 SPF grade male SD rats were randomly divided into the blank group,the model group,and the electro-acupuncutre(EA)group,with 12 rats in each group.The hepatic fibrosis model was established by CCL4 olive oil solution.The treatment group received electrical stimulation treatment on ST36,BL18,SP6,RN4 and LR3 one day after modeling,with each intervention lasting 15 minutes,once a day for 21 consecutive days.After the completion of modeling and 21 days of intervention,samples were taken from the liver tissues of rats in each group.The pathological changes and degree of liver fibrosis were observed using HE staining and Masson staining.ELISA method was used to detect the serum levels of hyaluronic acid(HA),laminin(LN),and type Ⅲ procollagen(PC Ⅲ).Real time fluorescence quantitative PCR was used to detect the relative expressions ofα-SMA and Collagen1 gene mRNA in liver tissues.Results:HE staining and Masson staining showed that a large number of liver cells occurred in the model group,and there were accumulations of necrosis and collagen fiber accumulation.In the treatment group,collagen fiber accumulation decreased and the liver cell structure tended to be normal.Compared with those in the blank group,the ELISA method detected that the serum levels of HA,LN and PC Ⅲ significantly increased in the model group;compared with those in the model group,the serum levels of HA,LN and PC Ⅲ decreased in the EA group.Real time fluorescence quantitative PCR experiment validated the contents ofα-SMA and Collagen1 mRNA increased(P<0.05),which proved that the model was successfully established,and the contents ofα-SMA and Collagen1 mRNA significantly decreased in the treatment group(P<0.05).Conclusion:Electro-needling ST36,GL18,SP6,RN4,and LR3 has a certain protective effect on hepatic fibrosis damage.
