摘 要:目的:探讨针刺“百会”透“曲鬓”通过JAK2/STAT3信号通路对脑出血模型大鼠的保护机制。方法:采用自体血注入尾壳核制取脑出血大鼠模型,将72只大鼠随机分为假手术组、模型组和针刺组,每组随机分为6 h、1 d、3 d和7 d共4个亚组。使用Berderson及Rosenberg评分法对各组大鼠神经功能缺损体征进行评价。HE染色观察脑组织形态学变化。Western blot法检测p-JAK2及p-STAT3表达水平。TUNEL法检测细胞凋亡率。结果:1 d、3 d和7 d时,模型组较针刺组大鼠Rosenberg评分降低;7 d时,模型组较针刺组大鼠Berderson评分降低,差异具有统计学意义(P<0.05)。与模型组比较,针刺组各时间点p-JAK2、p-STAT3蛋白回调显著,差异具有统计学意义(P<0.05)。与模型组比较,针刺组各时间点坏死神经元细胞减少,脑组织细胞凋亡率下降,差异具有统计学意义(P<0.05)。结论:针刺“百会”透“曲鬓”可改善脑出血大鼠的神经功能及血肿周围脑组织的病理变化,减少神经细胞凋亡,其机制与抑制JAK2/STAT3通路的激活有关。Objective:To explore the protective mechanism of needling Baihui(DU20)through Qubin(GB7)in intracerebral hemorrhage(ICH)rats via JAK2/STAT3 signaling pathway.Methods:A total of 72 rats with ICH were randomly divided into the sham operation group,the model group and the acupuncture group,each group was then randomly subdivided into 4 subgroups of 6h,1 d,3 d and 7 d.Berderson and Rosenberg scores were used to evaluate the signs of neurological impairment in each group.The morphological changes of brain tissues were observed by HE staining.The expressions of p-JAK2 and p-STAT3 were detected by Western blot.The apoptosis rate was detected by TUNEL method.Results:At 1 d,3 d and 7 d,the Rosenberg score of the model group was lower than that of the acupuncture group(P<0.05);at 7 d,Berderson score of the model group was lower than that of the acupuncture group(P<0.05).Compared with those in the model group,the protein rollbacks of P-JAK2 and P-STAT3 were significant at each time point in the acupuncture group(P<0.05).Compared with those in the model group,the necrotic neurons and the apoptosis rate of brain cells were both decreased in the acupuncture group at each time point(P<0.05).Conclusion:Needling DU20 through GB7 can improve the nerve function and the pathological changes of brain tissues around hematoma and reduce the apoptosis of nerve cells in rats with ICH,which mechanism is related to inhibiting the activation of JAK2/STAT3 pathway.
