摘 要:目的探讨宫颈癌患者人乳头瘤病毒(HPV)感染对癌组织人类Runt相关转录因子3(RUNX3)、酵母基因Noblp人类同源基因(NOB1)表达水平的影响。方法选取2021年2月至2023年10月在濮阳油田总医院行手术治疗的97例HPV感染的宫颈癌患者,另选取本院宫颈上皮内瘤变行手术治疗的30例患者,基于基因扩增技术及导流杂交原理,通过反向点杂交检测扩增产物与包被有型特异性探针膜杂交结果,采用碱性磷酸酶系统定性检测HPV感染状态,通过实时监测聚合酶链反应检测组织中RUNX3 mRNA、NOB1 mRNA表达水平,探讨病毒型别、病毒载量与组织中RUNX3、NOB1表达水平的关系。结果宫颈癌RUNX3 mRNA表达水平低于宫颈上皮内瘤变组织,NOB1 mRNA表达水平高于宫颈上皮内瘤变组织(P<0.05);随着FIGO分期增加、出现淋巴结转移患者NOB1 mRNA表达水平增加,RUNX3 mRNA表达水平降低,差异有统计学意义(P<0.05);宫颈癌患者中高危型HPV感染患者RUNX3 mRNA表达水平低于低危型别,差异有统计学意义(P<0.05),NOB1 mRNA表达水平高于低危型别,差异有统计学意义(P<0.05)。高HPV病毒载量宫颈癌患者RUNX3 mRNA表达水平低于低病毒载量,NOB1 mRNA表达水平高于低病毒载量(P<0.05);Spearman相关性分析结果显示RUNX3 mRNA表达水平与病毒载量呈现负相关(r=-0.407,P<0.005),NOB1 mRNA表达水平与病毒载量呈现正相关(r=0.392,P<0.005)。Logistic分析结果显示,高FIGO分期、淋巴结转移、高危型HPV感染、高病毒载量是RUNX3、NOB1高表达的影响因素。结论HPV感染宫颈癌患者癌组织NOB1高表达、RUNX3低表达与疾病分期及淋巴结转移相关,且HPV感染型别、病毒载量可影响癌组织NOB1、RUNX3表达水平。Objective To investigate the influence of human papillomavirus(HPV)infection on the expression levels of human Runt-related transcription factor 3(RUNX3)and yeast gene Nob1 homolog(NOB1)in cervical cancer tissues of patients.Methods A total of 97 HPV-infected cervical cancer patients who underwent surgical treatment at Puyang Oilfield General Hospital from February 2021 to October 2023 were selected for the study.Additionally,30 patients with cervical intraepithelial neoplasia who underwent surgical treatment at the same hospital were selected as a control group.Based on gene amplification technique and the principle of flow-through hybridization,the results of hybridization between the amplified product and coated specific probe membrane were detected by reverse dot blot,and the status of HPV infection was qualitatively detected by alkaline phosphatase system.The expression levels of RUNX3 mRNA and NOB1 mRNA in the tissues were monitored in real-time using polymerase chain reaction to explore the relationship between virus type,viral load,and the expression levels of RUNX3 and NOB1.Results The expression level of RUNX3 mRNA in cervical cancer tissues was lower than that in cervical intraepithelial neoplasia tissues,while the expression level of NOB1 mRNA was higher than that in cervical intraepithelial neoplasia tissues(P<0.05).As the FIGO stage increased and with the occurrence of lymph node metastasis,the expression level of NOB1 mRNA increased and RUNX3 mRNA expression decreased,showing statistically significant differences(P<0.05).In patients with cervical cancer,those infected with high-risk HPV had lower RUNX3 mRNA expression levels than those with low-risk types,and the difference was statistically significant(P<0.05).Conversely,NOB1 mRNA expression levels were higher in patients with high-risk HPV infection than in those with low-risk types,with statistically significant differences(P<0.05).In patients with high HPV viral load,RUNX3 mRNA expression levels were lower than those with low viral loads,while
