千斤拔不同提取物对胶原诱导性关节炎大鼠的影响

摘　　要：目的:探讨千斤拔不同提取物对胶原诱导性类风湿关节炎大鼠的影响。方法:建立胶原诱导性关节炎(collagen induced arthritis,CIA)大鼠模型,将造模成功的大鼠随机分为CIA组、水提取物组、石油醚组、乙酸乙酯组、正丁醇组,每组6只,另取6只正常大鼠为空白对照组。给药组通过灌胃给予相应药物,空白对照组与CIA组给予相应体积生理盐水,每天1次,连续14 d。实验过程中对各组大鼠进行关节炎指数评分及踝关节肿胀度测量,应用酶联免疫吸附法测定大鼠血清炎性因子水平,同时观察脏器指数的变化。结果:千斤拔水提取物、乙酸乙酯提取物、正丁醇提取物能够显著降低CIA大鼠关节炎指数及关节肿胀度(P<0.05,P<0.01),降低血清IL-1β、IL-6含量(P<0.05,P<0.01),而石油醚提取物无明显作用。结论:千斤拔水提取物、乙酸乙酯提取物、正丁醇提取物可减轻类风湿关节炎大鼠炎症症状,其作用机制可能与抑制炎症因子IL-1β、IL-6的释放有关。Objective To investigate the effects of different extracts of Moghania philippinensis Merr.on rheumatoid arthritis rats.Methods Collagen induced arthritis(CIA)model was established and the successfully modeled rats were randomly divided into 5 groups with random number:CIA group,water extract group,petroleum ether group,ethylacetate group and butanol group,each with 6 rats.In addition,6 normal rats were taken as blank control group.The rats in the administration group were given the drugs by intragastric administration,and the rats in the blank control group and the CIA group were given the corresponding volume of normal saline,once a day for 14 days.During the experiment,the arthritis index score and ankle swelling degree of rats in each group were measured.After the administration,the serum inflammatory factor levels of rats were determined by enzyme-linked immunosorbent assay(ELISA),and the changes of organ index were observed.Results The water extract,ethyl acetate extract and butanol extract of Moghania philippinensis could significantly reduce the arthritis index and joint swelling degree of CIA rats(P<0.05,P<0.01),and reduce the contents of IL-1βand IL-6 in serum(P<0.05,P<0.01),while the petroleum ether extract had no significant effect.Conclusion The water extract,ethyl acetate extract and butanol extract of moghania philippinensis extract can reduce the inflammatory symptoms of rheumatoid arthritis rats,and its mechanism may be related to the inhibition of the release of inflammatory factors IL-1βand IL-6.
