补骨脂素通过NF-κB途径抑制骨肉瘤细胞增殖的研究

摘  要:目的探索补骨脂素抑制骨肉瘤细胞增殖的分子机制。方法通过GeneCards、NCBI、OMIM、DisGeNET数据库获取骨肉瘤相关疾病基因,将从SwissTargetPrediction、SuperPred和SEA平台查询到的补骨脂素作用靶点和骨肉瘤相关靶点取交集筛选关键基因,然后用AutoDockTools进行分子对接,最后用骨肉瘤U2OS细胞验证分析结果。结果通过对不同数据库筛选出的靶点取交集共获得一个关键靶点NF-κB,分子对接结果显示补骨脂素与NF-κB稳定结合,CCK8结果显示补骨脂素能够显著抑制骨肉瘤细胞的增殖并呈现时间和剂量依赖性,免疫印记结果显示随着补骨脂素药物浓度的增加骨肉瘤U2OS细胞NF-κB表达量持续降低。结论本研究发现补骨脂素抑制骨肉瘤细胞增殖的作用可能与调控NF-κB有关。Objective To study the mechanism of psoralen inhibiting the proliferation of osteosarcoma cells.Methods Osteosarcoma-related disease genes were obtained from GeneCards,NCBI,OMIM and Dis-GeNET databases,and the key genes were screened by the intersection of psoralen targets and osteosarcoma-re-lated targets queried by Swiss Target Prediction,Super Pred and sea platforms.AutoDock was then used for molecular docking,and finally osteosarcoma U2OS cells were used to verify the results.Results A key target NF-κB was obtained by crossing the targets screened from different databases.Molecular docking results showed that psoralen could stably bind to NF-κB.CCK8 results showed that psoralen could significantly inhibit the proliferation of osteosarcoma cells in a time-and dose-dependent manner.Western blotting showed that the expression of NF-κB in osteosarcoma U2OS cells decreased with the increase of psoralen concentration.ConclusionThe inhibitory effect of psoralen on the proliferation of osteosarcoma cells may be related to the regulation of NF-κB.
