摘 要:目的:探究黄酮类甘草甙对慢性肾病大鼠血管钙化、肾间质炎症及NLRP3/caspase-1/IL-1β蛋白的影响。方法:40只大鼠分为正常(CO)组,模型(MO)组,低剂量甘草甙(LL)组,高剂量甘草甙(HL)组,每组10只,对MO组、LL组、HL组用腺嘌呤灌胃法建立慢性肾病大鼠模型,CO组不建模,建模成功后对LL组灌胃50 mg/kg甘草甙,对HL组灌胃150 mg/kg的甘草甙,CO组、MO组同期灌胃同体积生理盐水,HE染色检测肾组织病理形态,Von Kossa染色检测腹主动脉组织钙化情况,全自动生化分析仪检测血清生化指标,酶联免疫吸附法检测肾组织炎症相关因子,免疫印迹法及Real-time PCR法检测NLRP3/caspase-1/IL-1β表达。结果:CO组腹主动脉未见钙盐沉积,MO组腹主动脉可见大量黑色颗粒,与MO组比较,LL组、HL组腹主动脉黑色颗粒明显减少;与CO组相比,MO组Scr、BUN、P^(3-)、MCP-1、TNF-α、iNOS、NLRP3、caspase-1、IL-1β升高(P<0.05),Ca^(2+)降低(P<0.05);与MO组比较,LL组、HL组Scr、BUN、P^(3-)、MCP-1、TNF-α、iNOS、NLRP3、caspase-1、IL-1β降低(P<0.05),Ca^(2+)水平升高(P<0.05),且HL组比LL组变化明显(P<0.05)。结论:甘草甙对慢性肾病大鼠具有显著疗效,可显著改善血管钙化,降低肾间质炎症及NLRP3/caspase-1/IL-1β蛋白表达。Objective:To investigate the effects of flavonoid liquiritin on vascular calcification,renal interstitial inflammation and NLRP3/caspase-1/IL-1βprotein in rats with chronic kidney disease.Methods:Forty rats were divided into normal(CO)group,model(MO)group,low-dose liquiritin(LL)group and high-dose liquiritin(HL)group,with 10 rats in each group.The chronic kidney disease rat model was established by adenine gavage in MO,LL and HL groups,while no model was established in CO group.After successful modeling,LL group was gavage with 50 mg/kg liquiritin.The HL group was given 150 mg/kg liquiritin,the CO group and the MO group were given the same volume of normal saline at the same time.The pathological morphology of renal tissue was detected by HE staining,the calcification of abdominal aorta tissue was detected by Von Kossa staining,serum biochemical indexes were detected by automatic biochemical analyzer,and factors related to renal tissue inflammation were detected by enzyme-linked immunosorb method.The NLRP3/caspase-1/IL-1βexpression was detected by Western blot and Real-time PCR.Results:No calcium salt deposition was observed in the abdominal aorta of the CO group,and a large number of black particles were observed in the abdominal aorta of the MO group.Compared with the MO group,the black particles in the abdominal aorta of the LL and HL groups were significantly reduced.Compared with CO group,Scr,BUN,P,MCP-1,TNF-α,iNOS,NLRP3,caspase-1 and IL-1βin MO group were increased(P<0.05),and Ca was decreased(P<0.05).Scr,BUN,P,MCP-1,TNF-α,iNOS,NLRP3,caspase-1,IL-1βwere decreased in LL group and HL group(P<0.05),and Ca level was increased(P<0.05),and the changes in HL group were significantly higher than those in LL group(P<0.05).Conclusion:Liquiritin has a significant effect on chronic kidney disease in rats,which can significantly improve vascular calcification,reduce renal interstitial inflammation and NLRP3/caspase-1/IL-1βprotein expression.
