摘 要:为建立蒿柳组培繁育体系,以蒿柳茎段为外植体,采用改良WPM、MS、1/4 MS为基本培养基,研究不同植物生长调节剂种类及其浓度组合对腋芽萌发、不定芽增殖及生根的影响。结果表明:腋芽萌发以改良WPM+0.2 mg/L 6-BA+0.3mg/LNAA为优选培养基,腋芽诱导率可达90.21%,芽健壮、节间长,叶片鲜绿;继代培养中,添加1.0 mg/L的GA_(3)对促进不定芽的伸长生长、提高增殖系数有显著效果,芽增殖系数5.0;在改良WPM+0.5 mg/L IBA的培养基上,蒿柳继代苗生根率高且根粗壮、侧根多,生根率96.21%。To establish a tissue culture propagation system for Salix viminalis,stem segments of it were used as explants.An improved WPM,MS and 1/4 MS were employed as the basic culture medium.The effects of different types and concentrations of plant growth regulators on axillary bud germination,adventitious bud proliferation,and rooting were studies.The results showed that the optimum medium for axillary bud germination was the improved WPM+0.2mg/L 6-BA+0.3mg/LNAA,achieving a bud induction rate of 90.21%.The buds were robust,with elongated internodes and vibrant green leaves.In subculture,the addition of 1.0 mg/L GA_(3)significantly promoted the elongation and growth of adventitious buds and improved the proliferation coefficient to 5.0.On the improved WPM+0.5 mg/L IBA medium,the rooting rate of subcultured Salix shoots was high,with strong roots and numerous lateral roots,achieving a rooting rate of 96.21%.
