摘  要:【目的】探究脂多糖气管滴注对主流烟气诱导小鼠慢性阻塞性肺疾病模型的促进作用及中药HZSF的防治效果。【方法】选取C57BL/6J雄性小鼠40只,随机均分到空白对照组(CTL组)、卷烟烟气暴露组(CS组)、卷烟烟气暴露联合LPS气管滴注组(CS-LPS组)、中药HZSF对卷烟烟气暴露联合LPS气管滴注的干预组(HZSF组)。检测小鼠体重、肺脏功能、酚红排泌量、肺脏重量和肺脏组织病理学变化等指标。【结果】与CTL组相比,(1)CS组和CS-LPS组小鼠体重明显降低(P<0.05);HZSF组体重显著高于CS-LPS组(P<0.05);(2)CS组和CS-LPS组小鼠肺脏脏器系数显著升高(P<0.05);(3)CS组和CS-LPS组气道阻力明显升高(P<0.05),CS-LPS组第90ms用力呼气容积/用力呼气容积和呼气峰流量明显降低(P<0.05),HZSF组气道阻力和呼气峰流量则分别显著低于和高于CS-LPS组(P<0.05);(4)CS组和CS-LPS组的酚红排泌量明显升高(P<0.05),HZSF组的酚红排泌量则显著低于CS-LPS组(P<0.05);(5)出现肺泡腔扩张、肺泡壁断裂、炎症细胞浸润、杯状细胞增生和肺组织纤维化等病变,且CS-LPS组(10/10)病变比例显著高于CS组(6/10)和HZSF组(4/10)。【结论】LPS气管滴注对卷烟烟气暴露诱导小鼠COPD疾病模型有显著促进作用,但中药HZSF对小鼠COPD模型中的肺功能障碍和肺组织病变有明显改善效果。[Objective]This study aims to investigate the promoting effect of lipopolysaccharide(LPS)on a mouse chronic obstructive pulmonary disease(COPD)model induced by cigarette mainstream smoke(CS)and the prevention and treatment effects of a traditional Chinese medicine HZSF.[Methods]Forty C57BL/6J male mice were randomly divided into blank control group(CTL),cigarette smoke exposure group(CS),cigarette smoke exposure combined with LPS tracheal instillation group(CS-LPS),and traditional Chinese medicine HZSF intervention group for cigarette smoke exposure combined with LPS tracheal instillation(HZSF group).Indicators such as mouse body weight,lung function,phenol red secretion,lung weight,and histopathological changes in lung tissue were measured.[Results]①The body weights of CS group and CS-LPS group were significantly lower than that of CTL group(P<0.05).The body weight of HZSF group was significantly higher than that of CS-LPS group(P<0.05).②The coefficients of lung organ in CS group and CS-LPS group were significantly higher than those in CTL group(P<0.05).③For the measured lung functions,the airway resistances(RI)of CS group and CS-LPS group were significantly higher than those of CTL group(P<0.05).The RI and PEF of HZSF group was significantly lower and higher than that of CS-LPS group(P<0.05),respectively.The forced expiratory volume in 90 millisecond(FEV90)/the forced vital capacity(FVC)and peak expiratory flow(PEF)in CS-LPS group were significantly lower than those of CTL group(P<0.05).④Compared with CTL group,the amounts of sputum output in CS group and CS-LPS group were significantly higher than those of CTL group(P<0.05),and the amount of sputum output in the CS-LPS group was significantly higher than that of CS group(P<0.05),but the amount in the HZSF group was significantly lower than that of CS-LPS group(P<0.05).⑤The results of pathological staining of the lung tissue showed that the alveolar cavities were dilated,the alveolar walls were broken and fused into larger cavities.In addition,a large
