摘 要:通过质量平衡法和同位素稀释质谱法两种不同原理的方法对牛血清白蛋白进行纯度分析。采用液相色谱面积归一化法测量牛血清白蛋白的主成分纯度,顶空-气相色谱法测量挥发性有机成分,热重分析测定水分以及灼烧法测定灼烧残渣,最终确定牛血清白蛋白的纯度为0.962 g/g。采用基于氨基酸分析的同位素稀释质谱方法,通过苯丙氨酸与被同位素标记的苯丙氨酸之间的峰面积比值来最终确定牛血清白蛋白的纯度为0.964 g/g。两种方法对牛血清白蛋白定值分析结果有较好的一致性。Purity analysis of bovine serum albumin was conducted by using two methods based on different principles:mass balance method and isotope dilution mass spectrometry method.The main component purity of bovine serum albumin was measured by using liquid chromatography area nor-malization method.Volatile organic components were measured by using headspace gas chromatography method.The moisture was determined by thermogravimetric analysis method,and the ignition residue was determined by inustion method.The final purity of bovine serum albumin based on mass balance method was 0.962 g/g.Using the isotope dilution mass spectrometry method based on amino acid analysis,the purity of bovine serum albumin was ultimately determined to be 0.964 g/g by the peak area ratio between phenylalanine and isotope labeled phenylalanine.The two methods showed good con-sistency in the quantification results of bovine serum albumin.
