摘 要:[目的]探明乌梅对柱状黄杆菌的抑菌效果。[方法]将乌梅熬制至100 mg/mL的浓度,分别以1∶8、1∶16、1∶32、1∶64、1∶128、1∶256的比例加入液体培养基,接种柱状黄杆菌,培养72 h后观察并测定吸光度,进而判定抑菌效果;测定上述不同添加比例乌梅液体培养基的pH,并开展调整pH至7.2后的抑菌试验;分别以0.2、0.4、0.6 mg/mL的浓度加入固体培养基中,开展平板抑菌试验。[结果]未调节pH的液体药物培养基的最低抑菌添加比例为1∶128,即最低抑菌浓度为0.775 mg/mL。乌梅在添加比例高于1∶128时,对pH的影响较大,在浓度较低时pH的改变对抑菌效果的影响较小;在乌梅浓度为0.6 mg/mL时,可以对柱状黄杆菌完全抑菌。[结论]乌梅对柱状黄杆菌具有较好的抑菌效果;乌梅能够改变培养基的pH,但是在剂量较低时对抑菌试验结果的影响较小;乌梅对柱状黄杆菌的最低抑菌浓度为0.6 mg/mL。[Objective]To investigate the antibacterial effect of Fructus mume on Flavobacterium columnare.[Method]The antibacterial effect was determined by boiling Fructus mume to 100 mg/mL,adding the dosage of 1∶8,1∶16,1∶32,1∶64,1∶128 and 1∶256 into liquid medium,inoculating with F.columnare,observing and measuring absorbance after 72 h of culture.The pH of the liquid culture medium of Fructus mume with different addition ratios was determined,and the bacteriostatic test after pH to 7.2 adjustment was carried out.Then the concentration of 0.2,0.4 and 0.6 mg/mL were added into solid medium to carry out plate inhibition test.[Result]The minimum inhibitory addition ratio of unadjusted pH liquid drug culture medium was 1∶128,which means the minimum inhibitory concentration was 0.775 mg/mL.When the addition ratio of Fructus mume was higher than 1∶128,it had a significant impact on pH,and the effect of pH changed on antibacterial activity was relatively small at lower concentrations;when the concentration of Fructus mume was 0.6 mg/mL,it could completely inhibit the growth of F.columnare.[Conclusion]Fructus mume has a good antibacterial effect on F.columnare;Fructus mume can change the pH of the culture medium,but its effect on the antibacterial test results is relatively small at lower doses;the minimum inhibitory concentration of Fructus mume against F.columnare is 0.6 mg/mL.
