摘  要:目的:探究新疆伊犁黑蜂蜂胶提取物对粪肠球菌(E.f)浮游和生物膜状态的抗菌作用及在体内的有效性和安全性。方法:体外实验:采用液体稀释法测定最低抑菌浓度(MIC)和最低杀菌浓度(MBC),并测定生长曲线。通过结晶紫染色法测定最低生物膜抑制浓度(MBIC_(50))及最低已形成生物膜清除浓度(MBRC_(50))。利用SEM观察E.f生物膜表面形态。体内实验:建立大鼠根尖周炎动物模型并在用药前后采集大鼠根管内细菌行菌落计数测定;采集大鼠颌骨组织,通过Micro CT骨定量分析对比各组根尖周炎症的进展,记录实验期间各组大鼠的体重增幅,观察新疆伊犁黑蜂蜂胶提取物对大鼠生长发育的影响;取大鼠主要脏器进行HE染色,观察组织结构的改变,评估药物对大鼠主要脏器细胞的毒性。结果:体外实验:新疆伊犁黑蜂蜂胶提取物对E.f的MIC为0.938 mg/mL,MBC为1.875 mg/mL,MBIC_(50)为1.875 mg/mL,MBRC_(50)为3.75 mg/mL。SEM观察结果显示,与对照组相比,E.f生物膜形成量与蜂胶药液浓度成反比。体内实验:鼠龄10周,与空白组相比,蜂胶组大鼠根管内细菌量显著减少;蜂胶组及氢氧化钙组根尖周病变体积明显小于空白组,差异具有统计学意义(P<0.05)。各组大鼠体质量的生长趋势相似,主要脏器的HE染色切片均匀,组织学结构未见明显改变。结论:新疆伊犁黑蜂蜂胶提取物可安全有效地抑制E.f的生长及其生物膜的形成,为难治性根尖周炎(RAP)的治疗提供新的思路。AIM:To investigate the antibacterial effect of Xinjiang Yili black bee propolis extract on the planktonic and biofilm state of Enterococcus faecalis,as well as its efficacy and safety in vivo.METHODS:In vitro experiment:The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)were determined by liquid dilution method,and the growth curve was measured.The minimum biofilm inhibition concentration(MBIC_(50))and the minimum biofilm clearance concentration(MBRC_(50))were determined by crystal violet staining.The surface morphology of Enterococcus faecalis biofilm was observed by scanning electron microscopy(SEM).In vivo experiment:A rat model of periapical inflammation was established,and the bacteria in the root canal of the rats were collected and measured by colony count before and after medication.The development of periapical inflammation in each group was compared by Micro CT bone quantitative analysis.The weight increase of rats in each group was recorded during the experiment,and the effects of propolis extract of Xinjiang Yili on the growth and development of rats were observed.Major organs of the rats were subjected to hematoxylin and eosin(HE)staining to observe changes in tissue structure and evaluate the toxicity of the extract to the cells of major organs.RESULTS:In vitro experiment:The MIC and MBC of propolis extract on Enterococcus faecalis were 0.938mg/mL and 1.875 mg/mL respectively.The MBIC_(50) was 1.875 mg/mL and the MBRC_(50) was 3.75 mg/mL.The SEM results showed that the amount of Enterococcus faecalis biofilm formation was inversely proportional to the concentration of propolis liquid compared with the control group.In vivo experiment:Compared with the blank group,the bacteria quantity in the root canal of propolis group was significantly reduced at the age of 10 weeks;The periapical lesion volume of propolis group and calcium hydroxide group was significantly smaller than that of blank group,and the difference was statistically significant(P<0.05).The growth trend of
