摘 要:目的建立超高效液相色谱法(UPLC)测定贵州党参中党参炔苷含量,为贵州党参的质量控制提供方法参考。方法采用Waters CORTECS T_(3)色谱柱;流动相:乙腈-水(含0.1%磷酸)(23∶77);流速:0.4 ml/min;检测波长:268 nm。结果党参炔苷的线性方程Y=3.1963×104X+3.3047×10^(3)(r=0.9993);线性范围:0.01081~1.0808μg,党参炔苷的平均加样回收率为98.87%(n=6)。结论所建立的UPLC法测定贵州党参中党参炔苷含量操作快速简便,结果准确,重复性好,在实际工作中有较强的实用性,可用于贵州党参药材的质量控制。Objective To establish an UPLC method for the determination of lobetyolin in Guizhou Codonopsis Radix and to provide a method reference for the quality control of Guizhou Codonopsis Radix.Methods Waters CORTECS T_(3) chromatographic column was used.Mobile phase:acetonitrile-water(containing 0.1%phosphoric acid)(23∶77),flow rate:0.4 ml/min;the detection wavelength was 268 nm.Results The linear equation of lobetyolin was Y=3.1963×104X+3.3047×10^(3)(r=0.9993).Linear range:0.01081~1.0808μg,the average recovery of lobetyolin was 98.87%(n=6).Conclusion The established UPLC method was used to determine the content of lobetyolin in Guizhou Codonopsis Radix.The operation was fast and simple,the results were accurate,and the repeatability was good.It has strong practicability in practical work and can be used for quality control of Guizhou Codonopsis Radix.
