摘 要:目的探讨以五加皮对照提取物为对照的五加皮质量控制方法。方法采用高效液相色谱法(HPLC)检测,选用Waters Atlantis^(TM)T_(3)色谱柱(4.6×250 mm,5μm),以乙腈(A)-0.1%磷酸溶液(B)为流动相,梯度洗脱,以外标法计算五加皮中各组分的含量。结果新绿原酸、绿原酸、隐绿原酸、咖啡酸、1,3-O-二咖啡酰基奎宁酸、3,5-O-二咖啡酰基奎宁酸、4,5-O-二咖啡酰基奎宁酸、异贝壳杉烯酸的线性范围分别为0.2544~7.631、2.742~82.26、0.2165~6.494、0.04312~1.293、2.327~69.82、2.165~64.94、0.1840~5.520和4.456~133.7μg/ml(r≥0.9942),平均加样回收率范围为92.9%~102.7%,相对标准偏差(RSD)范围为0.90%~4.31%(n=6)。结论该方法简单、快速、经济,可用于五加皮中8种成分含量测定及五加皮质量控制。Objective To explore the quality control method of Acanthopanacis Cortex,extract Acanthopanacis Cortex were used as control.Methods The determination by High Performance Liquid Chromatography(HPLC)was performed on Waters Atlantis^(TM)T_(3) column with column temperature of 35℃.The acetonitrile(A)-0.1%phosphoric acid solution(B)was used as mobile phase by gradient elution.The content of each component in Acanthopanacis Cortex was calculated by external standard method.Results The linear ranges of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,caffeic acid,1,3-dicaffeioylquinic acid,3,5-dicaffeioylquinic acid,4,5-dicaffeioylquinic acid,and kaurenic acid were 0.2544~7.631、2.742~82.26、0.2165~6.494、0.04312~1.293、2.327~69.82、2.165~64.94、0.1840~5.520、4.456~133.7μg/ml(r≥0.9942)respectively.The recoveries rate were 92.9%~102.7%(RSD=0.90%~4.31%,n=6).Conclusion The method is simple,rapid and economical,and can be used for content determination of 8 components in Acanthopanacis Cortex.It can be used for quality control of Acanthopanacis Cortex.
