摘  要:精液冷冻保存的过程中精子细胞极易遭受损伤,其中过多的自由基会引起氧化应激,通过增加活性氧的产生对细胞系统产生负面影响。因此,精液冷冻保存中抗氧化剂的使用至关重要。迷迭香酸(rosmarinic acid,RA)是一种天然多酚类抗氧化剂,具有较强的抗氧化活性。本研究在奶山羊(Capra hircus)精液冷冻保存稀释液中添加0、20、40、80及120μmol/L迷迭香酸,检测精液质量及精子抗氧化能力,并通过LC-MS非靶向代谢组学技术分析了奶山羊精子冷冻后的精浆代谢特征。结果表明,迷迭香酸能够改善冷冻保存奶山羊精液的质量、提高精子抗氧化能力,在对照组与迷迭香酸处理组中检测到309种差异代谢物质,且迷迭香酸通过腺苷酸激活蛋白激酶(AMP-activated protein kinase,AMPK)信号通路调控精子的能量代谢,通过PI3K-Akt信号通路,激活或抑制B淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)等凋亡相关蛋白。研究表明,添加40μmol/L迷迭香酸能够改善奶山羊精液冷冻保存效果。本研究为优化奶山羊精液冷冻保存技术提供了新的思路与理论依据。Sperm cells are highly susceptible to damage during semen cryopreservation,in which excessive free radicals cause oxidative stress that negatively affects the cellular system by increasing the production of reactive oxygen species.Therefore,the use of antioxidants in semen cryopreservation is crucial.Rosmarinic acid(RA)is a natural polyphenolic antioxidant with strong antioxidant activity.In this study,0,20,40,80 and 120μmol/L RA was added to the semen cryopreservation diluent of dairy goats(Capra hircus)to test the sperm quality and sperm antioxidant capacity,and sperm plasma metabolic profiles of the spermatozoa of dairy goats were analyzed by LC-MS non-targeted metabolomics after freezing.The results showed that RA improved the quality of cryopreserved dairy goat spermatozoa and sperm antioxidant capacity,and 309differential metabolites were detected between the control group and RA-treated group.Moreover,RA regulated the energy metabolism of spermatozoa through AMP-activated protein kinase(AMPK)signaling pathway,and activated or inhibited apoptosis-related proteins,such as B-cell lymphoma-2(Bcl-2),through PI3K-Akt signaling pathway.The present study demonstrated that the addition of 40μmol/L RA could improve the effect of semen cryopreservation in dairy goats.This study provides a new idea and theoretical basis for the optimization of semen cryopreservation technology in dairy goats.
