反复种植失败患者子宫内膜FOXO1和ENaC的差异表达及其相关性研究

摘　　要：目的探讨叉头框蛋白O1(FOXO1)与反复种植失败患者子宫内膜上皮钠离子通道(ENaC)的相关性及其对反复种植失败患者的影响。方法将行体外受精-胚胎移植(IVF-ET)助孕并经历反复种植失败的患者(20例)设为研究组,将第一周期新鲜胚胎移植获临床妊娠患者(24例)设为对照组。采用实时荧光定量PCR法检测各组子宫内膜中α-ENaC mRNA表达水平,采用免疫印迹法检测各组患者子宫内膜中α-ENaC及磷酸化叉头框蛋白O1(pFOXO1)蛋白表达水平,分析2组蛋白在反复种植失败患者子宫内膜中表达及二者相关关系。结果反复种植失败组患者子宫内膜中α-ENaC mRNA及蛋白表达水平均较对照组下降,且差异均有统计学意义(P<0.05);反复种植失败组患者子宫内膜中pFOXO1蛋白水平表达水平较对照组下降,且差异均有统计学意义(P<0.05);2组患者子宫内膜中α-ENaC蛋白水平与pFOXO1蛋白水平呈正相关(r=0.471,P<0.05)。结论FOXO1与ENaC表达水平可能与反复种植失败患者子宫内膜容受性相关。Objective To investigate the regulatory effect of forkhead box 01(FOXO1)on epithelial sodium channel(ENaC)in patients with repeated implantation failure.Methods 20 patients who underwent in vitro fertilization-embryo transfer(IVF-ET)and experienced repeated implantation failures in the study group,24 patients with clinical pregnancy after the first cycle of fresh embryo transfer in the control group.The expression level ofα-ENaCmRNA in the endometrium of each group was detected by quantitative real-time PCR.The expression level ofα-ENaC protein and phosphorylation forkhead box O1(pFOXO1)protein in the endometrium of each group were detected by western blot.Results The mRNA and protein expression levels ofα-ENaC in endometrium of repeated implantation failure group were decreased compared with those of control group,and the differences were statistically significant(P<0.05).The expression level of pFOX01 protein in endometrium of repeated implantation failure group was decreased compared with that of control group(P<0.05).There was a positive correlation betweenα-ENaC protein and pFOXO1 protein level in endometrium of two groups(r=0.471,P<0.05).Conclusion ENaC and FOXO1 may be related to endometrial receptivity in patients with repeated implantation failure.
