摘  要:为明确引起湖北雪茄烟草根腐病的病原,在湖北省雪茄烟草主产区丹江口市、恩施州和宜昌市采集根腐病样品,采用常规组织分离法分离病原菌,通过室内带菌土壤接种验证其致病性,并进行了形态学和分子生物学鉴定。结果表明,镰刀菌在PDA培养基上菌落为白色至淡粉色,后期部分颜色加深,菌丝呈绒状。分生孢子呈典型的镰刀状,大型分生孢子多隔、无色、基本为镰刀形;小型分生孢子大部分无隔,呈椭圆形或纺锤形。基于ITS、EF-1α序列的系统发育分析,鉴定出引起湖北雪茄烟草根腐病的主要病原为尖孢镰刀菌(Fusariumoxysporum)和茄病镰刀菌(Fusariumsolani)。In order to clarify the pathogen causing root rot in Hubei cigar tobacco,diseased plant samples were collected from Danjiangkou,Enshi and Yichang regions,which are the main producing areas of Hubei cigar tobacco.The pathogen was isolated by conventional tissue isolation method,and its pathogenicity was verified by indoor inoculation of soil containing pathogen,and morphological identification and molecular biological identification were carried out.The results showed that the colonies of Fusarium on PDA medium were white to flesh-pink,and in some of them,the color deepened at a later stage and the mycelium was floccose.The conidia were typically sickle-shaped,the macroconidia were mostly septate,colorless,and sickle-shaped.Most of the microconidia were aseptate,elliptic or fusiform.Phylogenetic analysis based on ITS and EF-1αsequences identified Fusariumoxysporum and Fusariumsolani as the main pathogens causing root rot in Hubei cigar tobacco.
