摘 要:目的探讨五味子-黄芪混合多糖(Polysaccharide from mixed Schisandra chinensis and Astragali Radix,SAP)对D-半乳糖诱导小鼠肝损伤的保护作用及可能机制。方法KM小鼠随机分成5组,分别为空白组,模型组,SAP低(75 mg/kg)、中(150 mg/kg)、高(300 mg/kg)剂量组;颈背部皮下注射D-半乳糖(500 mg/kg)建立小鼠肝损伤模型。试验期间记录小鼠一般状态,测定小鼠肝脏指数,检测小鼠血清中ALT和AST水平、血清和肝脏中T-SOD活力和MDA含量,苏木精-伊红染色观察肝脏病理变化。结果与空白组比较,模型组小鼠肝脏指数、血清中ALT和AST水平显著升高(P<0.05,P<0.01);血清和肝脏中T-SOD活力均降低、MDA含量均升高(P<0.01,P<0.05);苏木精-伊红染色可知,小鼠肝细胞排列紊乱,细胞体积明显增大,胞浆疏松淡染,水样变性。与模型组比较,SAP高剂量组小鼠肝脏指数、血清中ALT和AST水平明显降低(P<0.05,P<0.01);血清和肝脏中T-SOD活力均有不同程度回升、MDA含量明显降低(P<0.01,P<0.05)。苏木精-伊红染色可知,各给药组肝脏病理受损程度明显减轻,肝细胞排列较为整齐,肝细胞肿胀、变形坏死程度减轻,水样变性得到改善。结论SAP对D-半乳糖诱导的肝损伤具有保护作用。Objective To investigate the protective effect and possible mechanism of action of polysaccharide from mixed Schisandra chinensis and Astragali Radix(SAP)against D-galactose-induced liver injury in mice.Methods KM mice were randomly divided into five groups:control group,model group,and SAP low(75 mg/kg),medium(150 mg/kg)and high(300 mg/kg)dose.The back of neck was injected subcutaneously with D-galactose(500 mg/kg)to establish a mouse liver injury model.During the test period,the general state of mice was recorded,the liver index was measured,and levels of ALT and AST in serum,and T-SOD activity and MDA content in both serum and liver were detected.Hematoxylin-eosin staining was used to observe pathological changes in the liver.Results Compared with control group,the liver index and serum levels of ALT and AST in model group were significantly higher(P<0.05,P<0.01).The T-SOD activity in both serum and liver was reduced,and the MDA content was elevated(P<0.01,P<0.05).Hematoxylin-eosin staining showed that the arrangement of hepatocytes was disorganized,with a marked increase in cell volume,and that the cytoplasm was sparsely and faintly stained,indicating hydropic degeneration.Compared with model group,the liver index and serum levels of ALT and AST in mice from high dose group of SAP were significantly reduced(P<0.05,P<0.01).T-SOD activity in serum and liver rebounded to varying degrees,and the MDA content was significantly reduced(P<0.01,P<0.05).Hematoxylin-eosin staining indicated that the degree of hepatic pathological damage was significantly reduced in each treated group,with hepatocytes arranged more neatly.Additionally,the swelling,deformation and necrosis of hepatocytes were reduced,and watery degeneration was improved.Conclusion SAP has a better protective effect on D-galactose-induced liver injury.
