摘  要:多胺氧化酶(PAO)是生物多胺分解代谢中的关键酶之一,为研究玛瑙红多胺氧化酶基因进化机制及其对非生物胁迫(高温,干旱)响应的特性,本研究以玛瑙红樱桃成熟叶片为材料,利用实验室构建的玛瑙红转录组数据库,筛选出PAO基因cDNA序列,对其开放阅读框(ORF)进行克隆和生物信息学分析。结果表明:克隆获得该基因cDNA全长为1917 bp;开放阅读框为1485 bp,编码494个氨基酸,预测蛋白大小为54.4 kD,等电点为5.28,为亲水性蛋白。RT-qPCR结果表明,CpPAO在不同组织中表达量存在差异,表达量依次为:膨大果(盛花期后45 d)>成熟果(盛花期后55 d)>初果(盛花期后35 d)>花芽>花>根>叶>茎。CpPAO能够响应高温、干旱胁迫,在胁迫处理后的叶片中,表达量明显提高。本研究为进一步解析CpPAO的功能奠定了理论基础。Polyamine oxidase(PAO)is a key enzyme involved in polyamine catabolism in plants.Toexplore the evolutionary mechanism of polyamine oxidase gene and its response characteristics to abiotic stress(high temperature,drought).The mature leaves of Chinese cherry(Cerasus pseudocerasus Lindl.'Manaohong')were used as materials in this study.The PAO gene cDNA sequence was screened from the Chinese cherry transcriptome database,and its open reading frame(ORF)was cloned and analyzed.The results showed that the full length of the cloned cDNA of the gene was 1917 bp;and the ORF was 1485 bp,which encoded 494 amino acids,the predicted protein size was 54.4 kD,the isoelectric point was 5.28,and the encoded protein was a hydrophilic protein.RT-qPCR results showed that the relative expression rate of CpPAO was different in multiple tissues:expanded fruit(45 days after full bloom)>ripe fruit(55 days after full bloom)>first fruit(35 days after full bloom)>flower bud>flower>root>leaf>stem.Additionally,CpPAO can respond to high temperature and drought stress,and the expression level of the leaves after the stress treatment was significantly increased.This research had laid a theoretical foundation for further analysis of the functions of CpPAO.
