摘  要:本研究采用RT-PCR的方法从菊花‘粉地毯’(Chrysanthemum morifolium‘Fen Ditan’)中克隆了CmTCP2基因(登录号:OR545672),通过生物信息学、亚细胞定位和基因表达分析,探讨CmTCP2基因在菊花头状花序形态建成中的功能,为通过分子生物学方法丰富菊花花型奠定基础。结果表明:(1)菊花CmTCP2基因的编码区序列长度为1332 bp,共编码443个氨基酸,为亲水性不稳定蛋白,属CIN类TCP转录因子。(2)系统进化和多序列比对分析显示,CmTCP2与野菊(C.indicum)CiTCP2的氨基酸序列相似性最高。(3)亚细胞定位发现,CmTCP2主要定位于细胞核。(4)实时荧光定量分析结果显示,CmTCP2在叶片和花器官中表达水平较高,在平瓣型‘粉地毯’和平瓣托桂型‘铺地淡粉’的舌状花中表达模式一致,管状花中表达模式相反,推测其在菊花叶片发育和花发育中发挥作用,参与花瓣发育过程,可能与管状花花瓣伸长形成桂瓣有关。本研究结果为深入探究CmTCP2在菊花花型调控中的功能提供科学依据。In this study,CmTCP2(accession number:OR545672)was cloned from C.morifolium'Fen Ditan'by RT-PCR.After that,bioinformatics analysis,subcellular localization analysis and gene expression analysis were performed to explore the function of CmTCP2 in the morphogenesis of chrysanthemum capitulum,and lay the foundation for enriching chrysanthemum flower types through molecular biology methods.The results showed that:(1)The coding sequence of CmTCP2 was 1332 bp,encoding 443 amino acids.CmTCP2 is a hydrophilic unstable protein and belongs to CIN class TCP transcription factor.(2)Phylogenetic and multiple sequence alignment analysis showed that the amino acid sequence of CmTCP2 was closely related to CiTCP2 in C.indicum.(3)Subcellular localization revealed that CmTCP2 was mainly localized in the nucleus.(4)The results of RT-qPCR showed that the expression level of CmTCP2 was relatively high in leaves and flower organs.The expression pattern of CmTCP2 was consistent in the ray florets of flat type'Fen Ditan'and flat anemone type'Pudi Danfen',but opposite in the disc florets.It is speculated that CmTCP2 plays a role in leaf and flower development of chrysanthemum,participates in the process of petal development,and may be related to the elongation of tubular petals to form anemone type petals.The results of this study provide a scientific basis for further investigation of the function of CmTCP2 in the regulation of chrysanthemum flower type.
