摘  要:为探究核桃内果皮露仁性状形成机理及JrMYB83与JrMYB46转录因子在核桃内果皮木质素生物合成途径中的作用,本研究通过RT-PCR方法克隆获得JrMYB83与JrMYB46基因,并利用实时荧光定量PCR技术分析其表达模式。结果表明:(1)JrMYB83与JrMYB46基因分别包含一个795和666 bp的开放阅读框,各编码264和221个氨基酸,蛋白质的相对分子质量分别为29.51和25.41 kD;氨基酸序列N末端均包含2个高度保守SANT结构域,表明两个基因属于R2R3类转录因子;系统发育树表明,JrMYB蛋白和核桃与山核桃的亲缘关系密切。(2)实时荧光定量PCR结果显示,JrMYB83与JrMYB46在核桃内果皮不同发育阶段和不同组织中的表达存在显著差异,且JrMYB83基因硬化区域的表达呈“M”变化趋势,在花后78 d表达量达到最高;JrMYB46在硬化部分呈现出“N”变化趋势,并于花后92 d到达峰值。推测2个MYB基因可能在核桃内果皮发育过程中起着关键作用。初步明确两个转录因子的功能,为核桃育种以及内果皮发育研究提供参考依据。In order to explore the mechanism of walnut endocarp character formation and the role of JrMYB83 and JrMYB46 transcription factors in the lignin biosynthesis pathway of walnut endocarp.In this study,JrMYB83 and JrMYB46 genes were cloned by RT-PCR,and their expression patterns were analyzed by real-time fluorescent quantitative PCR.The results were as follows:(1)JrMYB83 and JrMYB46 genes contain an open reading frame of 795 and 666 bp,respectively,each encoding 264 and 221 amino acids,and the relative molecular masses of protein are 29.51 and 25.41 kD,respectively;both N-terminal amino acid sequences contained two highly conservative SANT domains,indicating that the two genes belonged to R2R3 transcription factor class.The phylogenetic tree indicated that JrMYB protein and walnut were closely related to Carya illinoinensis.(2)Real-time fluorescent quantitative PCR results showed that the expression levels of JrMYB83 and JrMYB46 in different development stages and different tissues of walnut endocarp were significantly different,and the expression of hardened region of JrMYB83 gene showed a"M"change trend,reaching the highest level at 78 days after anthesis.JrMYB46 showed a trend of"N"change in the hardened part and peaked at 92 days after anthesis.It was speculated that two MYB genes might play key roles in the endocarp development of walnut.The functions of these two transcription factors were preliminarily clarified to provide reference for breeding and endocarp development of walnut.
