高粱低氮相关miRNA的表达聚类及染色体定位分析

摘  要:为深入了解miRNA在高粱低氮胁迫反应中的调控作用,本试验对高粱幼苗进行短时(8 h)和长时(15 d)低氮胁迫处理,利用Solexa深度测序技术结合生物信息学分析方法,筛选低氮相关差异表达的miRNA,并对其表达量进行聚类分析。同时利用TBtools软件对差异表达的miRNAs进行染色体定位分析。结果显示:低氮胁迫下,高粱根系和叶片中均有大量特有的小分子RNA序列,说明低氮胁迫可诱导高粱中小分子RNA的表达改变。对差异表达的miRNA进行聚类分析,根系中miRNA的表达聚为5种类型,叶片中miRNA的表达聚为4种类型。107个差异表达miRNAs不均匀分布在不同的染色体上,其中4号染色体上分布的最多。本试验结果为探究高粱耐低氮机制及培育耐低氮高粱品种提供参考依据。In order to deeply understand the regulatory role of miRNA in Sorghum response to low nitrogen stress,sorghum seedlings were treated with short-term(8 h)and long-term(15 d)low nitrogen stress.Solexa deep sequencing technology and bioinformatics analysis method were used to screen low nitrogen related differentially expressed miRNAs,and their expression levels were clustered and analyzed.At the same time,TBtools software was used to analyze the chromosome localization of differentially expressed miRNAs.The results showed that there were a large number of unique small molecule RNA sequences in Sorghum roots and leaves under low nitrogen stress,indicating that low nitrogen stress could induce the expression changes of small molecule RNA in sorghum.Cluster analysis of differentially expressed miRNAs showed that miRNA expression in roots was clustered into 5 types,and miRNA expression in leaves was clustered into 4 types.107 differentially expressed miRNAs were unevenly distributed on different chromosomes,with the most distributed on chromosome 4.The results of this experiment provide a reference for exploring the mechanism of low nitrogen tolerance of sorghum and cultivating low nitrogen tolerant sorghum varieties.
