摘 要:WOX(WUSCHEL-related homeobox)转录因子广泛地参与到对植物的生长和发育过程的调节,在植物的生长发育中起到非常重要的作用。但其关于木薯的嫁接效应,目前还鲜见报道。本研究采用生物信息学方法鉴定出木薯WOX基因家族的18个成员,亚细胞定位表明其成员全部位于细胞核。对木薯、拟南芥、西红柿WOX进行系统发生进化树分析,发现WOX基因家族可分为WUS分支、中间进化支和古老进化支。多重序列比对显示木薯WOX基因家族分为4个保守结构域。以木薯‘华南9号’和‘华南102’互为接穗和砧木,交互嫁接,通过RT-qPCR检测木薯嫁接植株叶片中18个MeWOXs基因的表达水平,结果显示在第1年的嫁接植株中,大部分MeWOXs基因均表达,但随着嫁接时间的延长,MeWOXs基因的表达数量逐渐减少,只有MeWOX1、MeWOX4、MeWOX7和MeWOX12在持续表达,其中,MeWOX7表达水平最高,推测它们在嫁接愈合过程中起重要作用。本研究结果为木薯成功嫁接提供理论依据。WOX(WUSCHEL-related homeobox)transcription factors are widely involved in the regulation of plant growth and development,and play an important role in plant growth and development.However,their effects on cassava grafting have been less studied.In this study,18 members of cassava MeWOX gene family were identified using bioinformatic methods.Subcellular localization showed that all the MeWOX genes were located in the nucleus.Based on the phylogenetic tree of WOX from cassava,Arabidopsis and tomato,the WOX gene family could be divided into WUS branch,intermediate branch and ancient branch.Multiple sequence alignment showed that there were four conservative domains in cassava MeWOX gene family.The cassava'South China 9'(SC9)and'South China 102'(SC102)were used as scions and rootstocks for mutual grafting.The expression levels of 18 MeWOXs genes in the leaves of cassava grafted plants were detected using RT-qPCR.The results showed that most of the MeWOXs in the 1st year grafted plants were expressed,but with the extension of grafting time,the number of expressed MeWOXs genes decreased gradually,except for MeWOX1,MeWOX4,MeWOX7 and MeWOX12,and MeWOX7 had the highest expression level.It was speculated that these MeWOXs genes would play an important role in the grafting healing process.The result of this study provide theoretical basis for successful grafting of cassava.
