台湾红藜EMS突变体库的构建与初步表型鉴定

摘  要:为深入探究藜麦品种资源的丰富性与其内在的基因功能,利用甲基磺酸乙酯(ethyl methane sulfonate,EMS)诱变处理台湾红藜种子,并对M2和M3代群体家系进行表型统计与筛选。以台湾红藜种子为材料,采用不同浓度EMS作为诱变剂(0,0.2%,0.6%,1%)分别处理6、12和18 h,通过统计种子的出苗率,最终筛选出0.6%浓度的EMS处理18 h作为诱变的半致死剂量参数。采用该半致死剂量参数创建台湾红藜突变体库,观察并统计诱变群体的突变率。经诱变处理得到6000份M1单株,选取其中400份M1单株进行种植鉴定,后代共获得709份发生一个到多个性状变异的突变株,主要包括茎秆、叶、穗、种子颜色、生育期以及其他变异类型。最终建立了台湾红藜的EMS诱变体系,获得对生产有利的突变体表型,为藜麦基因功能研究以及遗传育种改良提供了材料基础。The study aimed to use EMS(Ethyl methane sulfonate)mutagenic technology to construct a quinoa mutant library which will provide diverse resources for quinoa breeding and gene function study.Chenopodium formosanum Koidz.seeds were treated with different concentrations of EMS for 6,12 and 18 hours,respectively.By the statistics of seed emergence rate,the semi-lethal dose parameters of mutagenesis were determined and used to establish the mutant library of Chenopodium formosanum Koidz..The mutation rate of the mutagenic population was observed and counted to assess the quality of the library.Results show that the semi-lethal dose parameter of Chenopodium formosanum Koidz.seeds induced by EMS was treated with 0.6%EMS concentration for 18 h.6000 M1 plants were obtained and 400 of them were selected for the identification of phenotypes related to agronomic traits.These agronomic traits contained stem,leaf,ear,the color of seed,growth period etc.Finally,a total of 709 mutants with one or more character variations were screened out.The EMS mutagenesis system of Chenopodium formosanum Koidz.that the study have established contains diverse phenotypes beneficial to production and provides the material basis for the study of gene function and genetic breeding improvement of quinoa.
