摘 要:为探究不同浓度镉处理对水稻幼苗代谢物的影响及代谢通路变化,本研究利用气相色谱质谱联用仪对5种不同浓度(1,3,5,10,15 mg/L)镉胁迫下的水稻幼苗进行代谢物定性定量分析,通过主成分分析(PCA)及正交偏最小二乘法判别分析(OPLS-DA)结合多元统计分析方法筛选出水稻耐镉差异代谢物。结果显示,水稻幼苗中共检测出48种主要代谢物。经OPLS-DA分析后,在5种浓度下分别筛选出30、33、32、33、34个产生显著变化的差异代谢物,有20个共同差异代谢物,注释到发生变化的代谢通路共26条,其中有7条代谢通路受到明显扰动,且均与氨基酸代谢、有机酸代谢及生物合成相关。本研究结果为进一步探究水稻耐镉机制提供理论基础。In order to explore the effects of different concentrations of cadmium on the metabolites of rice seedlings and the changes in metabolic pathways,this study used gas chromatography-mass spectrometry to perform qualitative and quantitative analysis of metabolites on rice seedlings under five different concentrations(1,3,5,10,15 mg/L)of cadmium.The differential metabolites of cadmium tolerance in rice were screened by principal component analysis(PCA)and partial least square discriminant analysis(OPLS-DA)combined with multivariate statistical analysis.The results showed that a total of 48 main metabolites were detected in rice seedlings.30,33,32,33 and 34 differential metabolites with significant changes were selected respectively by OPLS-DA analysis.There were 20 common differential metabolites and a total of 26 metabolic pathways were annotated to be altered,of which 7 were significantly perturbed and all were related to amino acid metabolism,organic acid metabolism and biosynthesis.The results of this study provide a theoretical basis for further exploring the mechanism of cadmium tolerance in rice.
