摘  要:萜类化合物是植物界中数量最多的天然代谢物,具有多种重要的生物学功能,萜类合成酶(terpenoid synthase,TPS)在萜类物质合成中起着关键作用。莲(Nelumbo nucifera)是中国重要的药食两用的植物,关于其萜类物质及TPS基因家族的研究相对较少,本研究利用生物信息学技术鉴定莲的TPS基因家族,并分析了其基因结构、保守结构域、理化性质、蛋白质结构、基因表达特性等。结果表明,莲的TPS家族共有8个成员,分别命名为NnTPS01~NnTPS08。NnTPSs分为4个亚族,分布在3条染色体上,且均含有1个TPS和1个TPP结构域。理化性质分析表明,莲的TPS家族成员编码的蛋白均为酸性亲水蛋白。二级结构预测显示莲TPS基因家族编码的蛋白均以α-螺旋和无规卷曲为主。有6个蛋白亚细胞定位于叶绿体。顺式作用元件分析发现NnTPSs含有多种生物与非生物胁迫的作用元件。基因表达特性分析显示有7个基因可以表达,有5个基因在叶中表达量出现了上调,并且发现大多数基因在茎中都不表达。本研究结果为后续研究莲TPS基因家族功能特性提供科学依据。Terpenoids are the mo st abundant natural metabolites in the plant kingdom and have a variety of important biological functions.Terpenoid synthase(TPS)plays a key role in the synthesis of terpenoids.Nelumbo nucifera is an important medicinal and edible plant in China.There are relatively few studies on its terpenoids and TPS gene family.In this study,bioinformatics technology used to identify the TPS gene family in Nelumbo nucifera,and analyzed its gene structures,conserved domains,physical and chemical properties,protein structure,and expression profiles,etc.The results showed that TPS gene family has 8 members in N.nucifera,named as NnTPS01~NnTPS08.NnTPSs are divided into four sub-families,and they are distributed on three chromosomes,and each contains 1 TPS and 1 TPP domain.The physicochemical analysis showed that the proteins encoded by the TPS family members were acidic hydrophilic proteins.The secondary structure prediction showed thatα-helix and random coil are the main structures of the proteins encoded by NnTPSs.There are 6 proteins subcellular locations on the chloroplast.Cis-acting element analyzed found that NnTPSs contain a variety of biotic and abiotic stress elements.Analysis of gene expression characteristics showed that 7 NnTPSs could be expressed,and 5 NnTPSs were up-regulated in leaves.Moreover,it was found that most genes were not expressed in stems.The results of this study are expected to provide a reference for further research on the functional characteristics of NnTPS gene family.
