摘 要:目的探讨清除血管性血友病因子(von Willebrand factor,VWF)的主要器官。方法本研究主要采用荧光标记的VWF嵌合体(fluorescently labeled VWF chimera,FP⁃VWF KI)小鼠模型,通过组胺刺激模拟内源性VWF的释放。免疫荧光技术用于检测VWF在肝脏和脾脏的分布。此外,通过向野生型小鼠注射Cy5标记的人血浆VWF(Cy5⁃labeled human plasma VWF,Cy5⁃pd⁃VWF)研究了外源性VWF在体内的分布情况。结果研究结果表明,组胺刺激后,释放的内源性VWF在脾脏中分布显著增强。与此同时,注射的外源性Cy5⁃pd⁃VWF在肝脏中的积聚更为明显。结论脾脏在小鼠内源性VWF的清除过程中发挥主导作用。Objective To explore the primary organ responsible for the clearance of von Willebrand factor(VWF).Methods The current study primarily utilizes a fluorescently labeled VWF chimera(FP⁃VWF KI)mouse model,simulating the release of endogenous VWF through histamine stimulation.Immunofluorescence techniques were employed to detect the distribution of VWF in the liver and spleen.Furthermore,the study investigates the distribution of exogenous VWF in the body by injecting Cy5⁃labeled human plasma VWF(Cy5⁃pd⁃VWF)into wild⁃type mice.Results Previous studies,primarily based on the input of exogenous VWF,had identified the liver as the main organ for VWF clearance.Our findings indicated that endogenous VWF released by histamine stimulation was more prominently distributed in the spleen,whereas exogenous Cy5⁃pd⁃VWF accumulated mainly in the liver.Conclusion The spleen plays a leading role in the clearance process of endogenous VWF in mice.
